基于生物信息学分析miR-622在甲状腺癌中的表达及预后

摘要 目的通过生物信息挖掘分析miR-622在甲状腺癌中的表达,探究其靶基因功能富集及通路分析及其对甲状腺癌预后的影响,为甲状腺癌的治疗提供新思路。方法通过miRBase及dbDEMC 3.0数据库分析miR-622的保守性及在肿瘤中的表达;使用Starbase3.0和CancerMIRNome数据库筛选出miR-622的表达差异具有统计学意义的肿瘤。同时分析miR-622在THCA中的表达。对miR-622靶基因进行GO功能及KEGG通路富集分析,对下游靶基因建立相互作用网络,并采用Cytoscape软件对蛋白相互作用网络进行Hub基因筛选。通过Kaplan-Meier Plotter数据库分析miR-622表达与THCA患者总生存期的关系,评估其对THCA患者的预后意义。结果miR-622序列在多个物种间保持一致,具有高度的保守性。数据分析显示,miR-622在甲状腺癌中表达下调,且miR-622的低表达与THCA患者的不良预后有显著的相关性(P<0.05)。功能富集结果显示,miR-622靶基因主要参与P53信号通路、结肠癌以及神经营养因子信号通路等,通过靶基因分析,得到10个关键基因。结论miR-622在THCA患者中的表达异常,且miR-622参与THCA的多个生物过程及信号转导过程,可作为THCA潜在生物学标志物。 Objective To analyze the expression of miR-622 in thyroid cancer by bioinformatics,and to explore the functional enrichment and pathway analysis of its target genes,and its impact on the prognosis of thyroid cancer,so as to provide new ideas for the treatment of thyroid cancer.Methods The conservation of miR-622 and its expression in tumors were analyzed by miRBase and dbDEMC3.0 databases.Starbase3.0 and CancerMIRNome databases were used to screen out the tumors with statistically significant differences in miR-622 expression.The expression of miR-622 in THCA was also analyzed.GO function and KEGG pathway enrichment analysis of miR-622 target genes were performed,and the interaction network of downstream target genes was established.Cytoscape software was used to screen the Hub genes of protein interaction network.The relationship between miR-622 expression and overall survival of THCA patients was analyzed by Kaplan-Meier Plotter database,and its prognostic significance for THCA patients was evaluated.Results The miR-622 sequence was consistent among multiple species and had a high degree of conservation.Data analysis showed that miR-622 was down-regulated in thyroid cancer,and the difference was statistically significant(P<0.05).Functional enrichment results showed that miR-622 target genes were mainly involved in P53 signaling pathway,colon cancer and neurotrophin receptor signaling pathway.Ten key genes were obtained by target gene analysis.Conclusions The expression of miR-622 is abnormal in THCA patients,and miR-622 is involved in multiple biological processes and signal transduction processes of THCA,which can be used as a potential biological marker of THCA.
