摘要
为研究太行铁线莲的最佳繁殖方式,建立组织培养体系,以太行铁线莲的种子和带芽茎段为试材,分别进行了不同激素处理、不同水温浸种处理和不同贮藏方式处理的种子萌发试验,并采用4因素3水平正交设计对其扦插繁殖技术进行研究。结果表明,太行铁线莲种子经50 mg/L 6-BA浸种处理24 h后发芽率最高,为59.33%;以插穗顶部蘸取清水后扦插在草炭∶珍珠岩∶蛭石=1∶1∶1上,有利于其生根;太行铁线莲带芽茎段以0.1%HgCl 2消毒7 min时,出芽率最高。腋芽诱导最佳培养基为:MS+1.0 mg/L 6-BA+0.05 mg/L NAA,不定芽增殖最佳培养基为:MS+2.0 mg/L 6-BA+0.05 mg/L NAA+1.0 mg/L KT。
In order to study the best propagation method and establish a tissue culture system for Clematis kirilowii,seeds and germinated stem segments of C.kirilowii were used as test materials for seed germination tests with different hormone treatments,different water temperature immersion treatments and different storage treatments,and a 4-factor 3-level orthogonal design was used to study the propagation technique of its cuttings.The results showed that the highest germination rate of 59.33%was achieved after the seeds of C.kirilowii were dipped at 50 mg/L 6-BA for 24 hours.The cuttings were dipped in water at the top of the spike and rooted on grass charcoal∶perlite∶vermiculite=1∶1∶1,and the highest germination rate was achieved when the sprouted stem segments of C.kirilowii were disinfected with 0.1%HgCl 2 for 7 minutes.The best medium for axillary shoot induction was MS+1.0 mg/L 6-BA+0.05 mg/L NAA,and the best medium for adventitious shoot proliferation was MS+2.0 mg/L 6-BA+0.05 mg/L NAA+1.0 mg/L KT.
