摘要
目的:评价不同剂量藏红花素对急进高原失血性休克大鼠氧化应激和炎性反应的影响。方法:复制急进高原失血性休克大鼠模型,采用随机数字法将其分为5组(n=6):C组、NaCl组、LC组、MC、HC组,并给予不同剂量藏红花素。观察藏红花素复苏时间段MAP、SpO_(2)变化及各组大鼠生存时间。采用ELISA法测定血清中氧化应激指标:过氧化物酶(MPO)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)和肿瘤坏死因子-α(TNF-α)、白介素(IL)-18、IL-6、IL-1β的含量。结果:藏红花素干预后MC组及HC组的急进高原失血性休克大鼠的存活时间及存活率均提高。与C组相比,其余四组在休克后(复苏时)、复苏0.5 h、复苏1 h的MAP均下降(P <0.05)。与NaCl组相比,MC组、HC组、C组在复苏0.5 h、复苏1 h的SpO_(2)均升高(P <0.05);与C组相比,其余四组的SpO_(2)在休克后(复苏时)、复苏0.5 h、复苏1 h均下降(P <0.05)。藏红花素呈剂量依赖性降低血清炎性因子TNF-α、IL-18、IL-6、IL-1β,同时藏红花素能明显升高休克大鼠血清中SOD、GSH活性及降低MPO、MDA的含量。结论:藏红花素可通过剂量依赖性抑制急进高原失血性休克大鼠的炎症反应和氧化应激来提高生存率。
Objective:The purpose of this study is to evaluate the effects of different doses of crocin on oxidative stress and inflammation in rats acute exposure to high altitude with hemorrhagic shock.Methods:The rat models of acute exposure to high altitude with hemorrhagic shock were replicated and different doses of crocin were given.The changes of MAP and SpO_(2) and survival time of each group of rats were observed during the corresponding time periods of crocin resuscitation,and the serum levels of oxidative stress indicators myeloperoxidase(MPO),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione(GSH) and inflammatory factors tumor necrosis factor-α(TNF-α),interleukin(IL-18,IL-6,IL-1β) were measured by ELISA.Results:MC and HC groups improved the survival time and survival rate of rats in acute exposure to high altitude with hemorrhagic shock after crocin intervention.Compared with group C,the MAP of the remaining four groups decreased after shock(at resuscitation),at 0.5 h and at 1 h of resuscitation(P<0.05).Compared with the NaCl group,the SpO_(2) in MC,HC and C groups at 0.5 h and 1 h of resuscitation was increased(P 0.05);and compared with the C group,the SpO_(2) in the remaining four groups decreased after shock(at resuscitation),0.5 h and 1 h of resuscitation(P<0.05).Crocin dose-dependently decreased serum inflammatory factors TNF-α,IL-18,IL-6 and IL-1β,while crocin significantly increased SOD and GSH activities and decreased MPO and MDA in the serum of shocked rats.Conclusion:Crocin may improve the survival rate by dose-dependent inhibition of inflammatory response and oxidative stress in rats acute exposure to high altitude with hemorrhagic shock.
