基于网络药理学和实验验证探究补阳还五汤治疗糖尿病的分子机制 被引量:6

Investigation of the molecular mechanism of Buyang Huanwu Decoction in the treatment of diabetes based on network pharmacology and experimental verification

摘要 目的:基于网络药理学、分子对接及动物实验验证补阳还五汤治疗糖尿病的物质基础及作用机制。方法:从TCMSP、GeneCards、OMIM、Venny获取糖尿病与补阳还五汤的交集靶点。用Cytoscape对“活性成分-靶标”网络进行拓扑分析。通过Metascape对共同靶点进行GO、KEGG富集分析,分子对接技术分析药物与靶点的结合能力。在糖尿病大鼠模型上检测影响血糖的相关指标及关键蛋白的表达情况。结果:共得到补阳还五汤中活性成分116个,“药物-疾病”交集靶点85个。关键成分包括槲皮素、山奈酚、木犀草素等。动物实验验证补阳还五汤可显著降低糖尿病大鼠的血糖、血脂,并提高Akt1、VEGFA、Caspase-3蛋白的表达。结论:补阳还五汤中的槲皮素、山奈酚、木犀草素等成分可通过Akt1、VEGFA、Caspase-3等蛋白激活Insulin、VEGF、PI3K/Akt等信号通路,调控大鼠糖脂代谢,从而实现多成分、多靶点、多通路影响糖尿病发生发展的目的。 OBJECTIVE To verify the material basis and mechanism of Buyang Huanwu Decoction in the treatment of diabetes based on network pharmacology,molecular docking and animal experiments.METHODS The intersection targets of diabetes and Buyang Huanwu Decoction were obtained from TCMSP,GeneCards,OMIM,and Venny.Topological analysis of the"active component-target"network was carried out by Cytascape.GO and KEGG enrichment analysis of common targets were carried out by Metascape,and the binding ability of drugs and targets was analyzed by molecular docking technology.The related indexes affecting blood glucose and the expression of key proteins in diabetic rat model were detected.RESULTS A total of 116 active ingredients and 85"drug-disease"intersection targets were obtained for Buyang Huanwu Decoction.Key ingredients included quercetin,kaempferol and luteolin,etc.Animal experiments proved that Buyang Huanwu Decoction significantly reduced blood sugar and blood lipid in diabetic rats,and increased the expression of Akt1,VEGFA,and Caspase-3 proteins.CONCLUSION Quercetin,kaempferol,luteolin and other components in Buyang Huanwu Decoction can activate insulin,VEGF,PI3K/Akt and other signal pathways through Akt1,VEGFA,Caspase-3 and other proteins to regulate glucose and lipid metabolism in rats,so as to achieve the goal of multi-component,multi-target,and multi-channel effects on the occurrence and development of diabetes.
