摘要
以沙棘叶为材料,分别以水、甲醇、乙醇为提取溶剂,采用超声波辅助法提取沙棘叶中多酚类化合物,并通过DPPH自由基和羟自由基清除率试验研究沙棘叶多酚的抗氧化活性。试验结果表明,60%乙醇作为提取溶剂时,沙棘叶多酚得率最高。不同溶剂提取的沙棘叶多酚得率顺序为:60%乙醇>60%甲醇>热水。抗氧化试验结果表明,沙棘叶多酚对DPPH自由基和羟自由基均有显著的清除能力。不同溶剂提取的沙棘叶多酚对DPPH和羟自由基的清除率不同,其中60%乙醇提取的多酚对DPPH、羟自由基的清除率最高,分别达98.2%、44.3%,明显优于阳性对照Vc,结果表明沙棘叶可以作为开发天然抗氧化剂的资源。
Polyphenols were extracted from Seabuckthorn leaves using water,methanol and ethanol as extraction solvents.Ultrasonic assisted method was used to extract polyphenols from Seabuckthorn leaves.The antioxidant activity of polyphenols from Seabuckthorn leaves was studied by DPPH radical scavenging and hydroxyl radical scavenging tests.The results showed that the yield of polyphenols from seabuckthorn leaves was the highest when 60%ethanol was used as the extraction solvent.The order of yield of polyphenols from Seabuckthorn leaves was 60%ethanol>60%methanol>hot water.The antioxidant test results showed that polyphenols from seabuckthorn leaf had significant scavenging ability on DPPH free radical and hydroxyl free radical.The scavenging rates of DPPH and hydroxyl free radical of polyphenols extracted from sea buckthorn leaves were different with different solvents.The scavenging rates of polyphenols extracted from 60%ethanol were the highest,reaching 98.2%and 44.3%,respectively,which were significantly better than the positive control VC.The results indicated that sea buckthorn leaves could be used as natural antioxidant resources.
