摘要
目的探讨温针对膝骨关节炎(KOA)模型大鼠行为表现及相关因子B细胞淋巴瘤-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)的影响。方法将24只SPF级大鼠随机分为空白组、模型组与温针组,每组8只。膝关节腔注射4%木瓜蛋白酶建立KOA大鼠模型,用温针对温针组进行干预,每次20 min,每日1次,干预6 d休息1 d,1周为1个疗程,共2个疗程。干预结束后,对大鼠进行Lequesne MG动物行为学评分,检测软骨组织内Bcl-2与Bax的mRNA表达。结果造模后,模型组与温针组行为学评分均明显高于空白组(P<0.01)。疗程结束后,与模型组相比,温针组行为学评分显著降低(P<0.01),Bcl-2 mRNA表达增加(P<0.05),Bax mRNA表达下降(P<0.05),差异均有统计学意义。结论温针可能通过提高Bcl-2的基因表达、抑制Bax mRNA的表达对膝关节软骨起保护作用,从而改善KOA模型大鼠的日常行为活动能力。
Objective To investigate the effects of warm needle moxibustion on behaviors,Bcl-2 and Bax of KOA model rats.Methods 24 SPF rats were randomly divided into normal,model and warm needle moxibustion(WNM)groups,8 per group.The KOA rat model was established by injecting 4 percent of papain into the joint cavity.The WNM group received warm needle moxibustion for 20 min once a day.1 week is a course of treatment,which intervene 6 days and 1 day off,and there were 2 courses in total.After treatment,using Lequesne MG to evaluate rats’behaviors,detecting the expressions of Bcl-2 mRNA and Bax mRNA in knee cartilage of rats by qPCR.Results After knee osteoarthritis model established,the scores of Lequesne MG of model group and WNM group were significantly higher than the normal group(P<0.01).The expression of Bcl-2 mRNA of model group decreasedgroup.After treatment,Compared to model group,the Lequesne MG score of WNM group significantly reduced(P<0.05),the expression of Bcl-2 mRNA increased,and the expression of Bax decreased,and the differences were considered statistically significant(P<0.05).Conclusion Warm needle moxibustion may protect the knee articular cartilage,by increasing the gene expression level of Bcl-2 and Bax,thereby improving the daily behaviors and activities of KOA model rats.
