摘要
目的探讨高压氧(HBO)联合雷公藤甲素(TPL)处理对大肠癌SW620细胞凋亡及相关信号通路的影响。方法体外培养大肠癌SW620细胞完成后,将细胞分为4组,对照组不作处理,TPL组加入终质量浓度为2 mg/L的TPL溶液,HBO组则在高压氧舱中暴露培养90 min,HBO+TPL组将SW620细胞先在高压氧舱暴露培养90 min后用2 mg/L的TPL溶液处理。采用CCK-8法检测SW620细胞的增殖能力,流式细胞术检测SW620细胞的凋亡情况,Western blotting实验检测SW620细胞凋亡相关蛋白及相关信号通路蛋白的表达水平。结果HBO+TPL组SW620细胞的存活被进一步抑制,其存活率[(52.13±2.43)%]明显低于TPL组[(78.54±2.47)%],差异有统计学意义(P<0.05)。HBO+TPL组SW620细胞凋亡率[(38.2±3.2)%]明显高于TPL组[(28.4±2.1)%],差异有统计学意义(P<0.05)。HBO+TPL组SW620细胞凋亡蛋白Bcl-2表达较TPL组明显降低,Bax和caspase-3的表达较TPL组明显升高(P<0.05)。与HBO组和对照组比较,TPL组SW620细胞p-JNK表达明显升高,p-STAT3表达明显降低(P<0.05);与TPL组比较,HBO+TPL组SW620细胞p-JNK表达明显升高、p-STAT3表达及耐药性相关蛋白P-gp表达明显降低(P<0.05)。结论HBO协同处理能够增强TPL对SW620细胞的增殖抑制作用及诱导凋亡作用,其作用机制可能是通过调节SW620细胞中的JNK/STAT3信号通路实现的。
Objective To investigate the effect of hyperbaric oxygen(HBO)combined with triptolide(TPL)on apoptosis and related signal pathways of colorectal cancer SW620 cells.Methods After being cultured in vitro,the SW620 cells were divided into four groups:the control group received no treatment;the TPL group was added with TPL solution with a final concentration of 2 mg/L;the HBO group was exposed in HBO chamber for 90 min;and the HBO+TPL group was exposed in HBO chamber for 90 min and then given 2 mg/L of TPL.The CCK-8 was used to detect the proliferation of SW620 cells.The flow cytometry was used to detect the apoptosis of SW620 cells.The western blotting was used to detect the expression of apoptosis-related proteins and signal pathway related proteins in SW620 cells.Results The survival of SW620 cells in HBO+TPL group was further inhibited,and its survival rate was significantly lower than that in the TPL group[(52.13±2.43)%vs.(78.54±2.47)%](P<0.05).The apoptosis rate of SW620 cells in the HBO+TPL group was significantly higher than that in the TPL group[(38.2±3.2)%vs.(28.4±2.1)%](P<0.05).The expression of apoptosis protein Bcl-2 in the HBO+TPL group was significantly lower than that in the TPL group,while the expressions of Bax and caspase-3 in the HBO+TPL group were significantly higher than those in the TPL group(P<0.05).Compared with the HBO group and the control group,the expression of protein p-JNK in SW620 cells in the TPL group increased and the expression of p-STAT3 decreased(P<0.05).Compared with the TPL group,the expression of protein p-JNK in the HBO+TPL group increased,the expression of p-STAT3 and drug resistance-related protein P-gp decreased(P<0.05).Conclusion HBO auxiliary treatment can effectively enhance the inhibitory effect of TPL on the proliferation of SW620 cells,as well as its regulatory effect on cell apoptosis,the mechanism of action of which may be achieved by regulating the JNK/STAT3 signaling pathway in SW620 cells.
