摘要
目的:研究牛樟叶总多糖减轻衰老小鼠认知障碍的作用。方法:6月龄60只昆明小鼠,随机分为空白组、模型组、阳性组盐酸多奈哌齐片(0.3 mg/100 g)、牛樟叶总多糖提取物给药组(58 mg/100 g、46.4 mg/100 g、29 mg/100 g)。采用腹腔注射D-半乳糖方法制备衰老动物模型,连续注射给药40 d,于第11天开始给予相应剂量的药物,第36天进行Morris水迷宫考察小鼠行为学实验,给药结束后,用水合氯醛麻醉后处死取材,采用酶标仪按试剂盒说明书检测脑匀浆丙二醛(MDA)水平、小鼠血清超氧化物歧化酶(SOD)、谷光甘肽(GSH)水平、谷胱甘肽过氧化物酶(GSH-Px),采用HE染色观察小鼠海马组织形态学改变情况。结果:与模型组相比,牛樟叶提取物各剂量组能不同程度降低小鼠脑匀浆中MDA水平,升高小鼠血清中SOD、GSH、GSH-Px水平,牛樟叶总多糖提取物组认知功能有所提高,与模型组比较,阳性药物组和牛樟叶总多糖各剂量组小鼠海马组织CA1区神经细胞排列较整齐,结构相对正常,核浓染和固缩的程度降低。结论:牛樟叶总多糖有一定的抗衰老减轻认知障碍的作用,作用机制可能与其抗氧化作用有关。
Objective To study the effect of total polysaccharide from cinnamomum cinnamomum leaves on reducing cognitive impairment in aged mice.Methods Sixty Kunming mice aged 6 months were randomly divided into blank group,model group,positive group,donepezil hydrochloride tablet(0.3 mg/100 g),and cinnamomum cinnamomum leaves polysaccharide extract administration groups(58 mg/100 g,46.4 mg/100 g,29 mg/100 g).The aging animal model was prepared by intraperitoneal injection of D-galactose for 40 days.On the 11th day,the corresponding dose of the drug was given.On the 36th day,the Morris water maze experiment was conducted to investigate the behavior of the mice.The level of malondialdehyde(MDA)in brain homogenate,the level of superoxide dismutase(SOD),glutamyl glycine(GSH)and glutathione peroxidase(GSH-Px)in serum of mice were detected by microplate plate according to the kit instruction.The morphological changes of hippocampus were observed by HE staining.Results Compared with the model group,each dose group of cinnamomum cinnamomum extract could reduce the level of MDA in rat brain homogenate to different degrees,and increase the levels of SOD,GSH and GSH-Px in serum of mice.The cognitive function of cinnamomum cinnamomum cinnamomum polysaccharide extract group was improved.The nerve cells in the hippocampal CA1 region of mice in the positive drug group and the total cinnamomum cinnamomum polysaccharide groups were arranged neatly and the structure was relatively normal,and the degree of nuclear hyperchromia and pyknosis were reduced.Conclusion Polysaccharide from Cinnamomum cinnamomum leaves has a certain anti-aging effect and alleviates cognitive impairment,and its mechanism may be related to its antioxidant effect.
