摘要
为明确用于食用菌粗多糖含量测定的苯酚-硫酸分光光度法中对结果可靠性的影响因素,根据标准NY∕T 1676—2008中的方法对一个灵芝破壁孢子粉样品的粗多糖进行测定,分析该方法在测定过程中的不确定度来源,计算各分量的不确定度和合成不确定度,并对各分量不确定度对合成不确定度的贡献大小进行排名。结果表明:灵芝破壁孢子粉样品的粗多糖含量为2.71%±0.09%(k=2),影响不确定度的主要来源是标准品溶液配制和稀释、标准曲线拟合、样品重复性。增加标准工作溶液的配制量可有效降低苯酚-硫酸分光光度法的不确定度,并能提高其可靠性。
In order to clarify the influencing factors of the results reliability in the phenol-sulfuric acid spectrophotometry used for the determination of the crude polysaccharide content of edible fungi,the crude polysaccharide of a Ganoderma lucidum broken spore powder sample was determined by the standard method of NY∕T 1676—2008,then the sources of uncertainty was analyzed,and the uncertainty of each component as well as the combined uncertainty were calculated.At last,the contribution of each component uncertainty to the combined uncertainty was ranked.The results showed that,the crude polysaccharide content of Ganoderma lucidum broken spore powder samples was 2.71%±0.09%(k=2),and the main sources of uncertainty were standard solution preparation and dilution,standard curve fitting,and sample repeatability.The uncertainty of the method could be effectively reduced by increasing the preparation amount of the standard working solution.
