摘要
目的食用菌中的多糖成分具有较高的药用保健价值,为了提高食用菌中多糖的提取效率,本研究对经典的食用菌多糖提取方法—热水浸提-醇沉淀法进行了优化。方法选取影响热水浸提-醇沉淀法提取效率的三个主要因素,即沸水浴加热时长,超声波提取时长和醇沉浓度分别进行优化。采用SPSS 21.0软件进行Wilcoxon符号秩和检验,检验水准α=0.05。结果通过条件优化实验可知,当沸水浴加热时长为1.5 h,超声波提取时长为15 min,醇沉浓度为70%时,食用菌中多糖提取效率最佳。优化后提取技术的相对标准偏差范围是1.15%~1.24%,加标回收率范围是89.5%~95.9%。将优化后的提取技术用于各种类食用菌样品中,两者检测结果经统计学检验得P<0.01,可认为优化后的方法比原方法具有更高的提取效率。结论本研究提高了食用菌粗多糖的提取效率,研究结果为进一步地开发和利用食用菌多糖提供了一定的参考依据。
Objective The polysaccharides in edible fungi have high values for medicine and health.In order to improve the extraction efficiency of polysaccharides in edible fungi,we aimed to study the extraction technology of crude polysaccharide from edible fungi.Methods Three main factors affecting the extraction efficiency of hot water extraction-alcohol precipitation method were selected,including the heating time of boiling water bath,the extraction time of ultrasonic wave and the concentration of alcohol precipitation.Then,those three main factors were optimized in this study.SPSS 21.0 software was used for wilcoxon signed rank sum test,and the test levelα=0.05.Results By optimizing the conditions,the extraction efficiency of polysaccharides from edible fungi achieved the best when the heating time of boiling water bath was 1.5 h,the extraction time of ultrasonic wave was 15 min and the concentration of alcohol precipitation was 70%.The relative standard deviation of the optimized extraction technology was 1.15%-1.24%,and the recovery rate ranged from 89.5% to 95.7%.The new extraction method was applied to edible fungi samples.The results of the two methods were compared,and we found the optimized method has higher extraction efficiency than the original method(P<0.01).Conclusion The extraction method improve the extraction efficiency of polysaccharides in edible fungi in this study.Our findings have provided a reference for further development and utilization of Polysaccharides from edible fungi.
