梓醇对大鼠急性脊髓损伤的神经保护作用及其机制研究 被引量:1

摘要 目的检测梓醇(Catalpol)对于大鼠急性脊髓损伤(ASCI)的自噬、凋亡相关蛋白及运动功能的影响,并研究梓醇对大鼠脊髓损伤的神经保护作用的机制。方法20只成年SD雄性大鼠,通过SPSS随机数生成法分为空白对照组(Sham组)和SCI模型组和低、中、高剂量梓醇组共5组,大鼠通过Allen′s法模拟脊髓损伤建模。术后7 d对各组大鼠进行BBB评分、脊髓组织形态观察、Bax及LC3蛋白含量检测,p62、LC3蛋白免疫组化染色。结果与Sham组相比,SCI组观测到大量坏死神经元细胞,BBB评分明显降低(P<0.001),LC3-Ⅱ、Bax明显增加(P<0.001),与SCI组相比,梓醇三个剂量组空泡状坏死神经元细胞明显减少,BBB评分明显增加(P<0.05),LC3-Ⅱ明显增加(P<0.05),Bax明显减少P<0.01),免疫组织化学染色p62、LC3的表达增强。结论梓醇能减少大鼠急性脊髓损伤后神经元细胞凋亡,促进运动功能恢复,机制可能与其增强细胞自噬来抑制细胞凋亡有关。 OBJECTIVE To investigate the effects of Catalpol on autophagy,apoptosis-related proteins and motor function in rats with acute spinal cord injury(ASCI),and to study the neuroprotective mechanism of catalpol on spinal cord injury in rats.METHODS Twenty adult SD male rats were randomly divided into blank control group(Sham group),SCI model group and low,medium and high dose catalpol group by SPSS.Rats were modeled by Allen′s method to simulate spinal cord injury.Seven days after operation,BBB score,observation of spinal cord tissue morphology,detection of Bax and LC3 protein content,and immunohistochemical staining of p62 and LC3 protein were performed in rats of each group.RESULTS Compared with Sham group,a large number of necrotic neurons were observed in SCI group,and the BBB score decreased significantly(P<0.001),while LC3-Ⅱand Bax increased significantly(P<0.001).Compared with SCI group,the number of vacuolar necrotic neurons in catalpol three dosage groups decreased significantly,the BBB score increased significantly(P<0.05),and LC3-Ⅱincreased significantly,while the level of Bax decreased significantly(P<0.01).CONCLUSION Catalpol can reduce neuronal apoptosis and promote the recovery of motor function after acute spinal cord injury in rats,and the mechanism may be related to its inhibition of apoptosis by enhancing autophagy.
