青叶胆多酚复合酶协同超声辅助提取工艺及抗氧化性研究

摘要 为确定复合酶协同超声辅助提取青叶胆多酚Swertia mileens polyphenols(SMP)的最佳工艺及其抗氧化活性,以青叶胆为材料,设计复合酶协同超声辅助提取工艺单因素试验,筛选影响显著因素,再利用Box-Benhnken中心组合设计响应面试验,确定最优工艺,并通过测定SMP对DPPH自由基(DPPH·)和亚硝酸根离子(NO2-)的清除能力来评价其抗氧化性。结果表明:复合酶协同超声辅助提取SMP最佳工艺条件为酶解温度40℃,酶解时间50 min,液料比35∶1 mL·g^(-1),乙醇体积分数63%,超声功率300 W,复合酶用量6%(木瓜酶用量5%,纤维素酶用量1%,以青叶胆干粉质量为基准,下同),在此条件下SMP提取量为17.50 mg·g^(-1);SMP对DPPH自由基、亚硝酸根离子有较强的清除能力,且随着多酚质量浓度的增大,清除能力增强;SMP质量浓度为0.007 mg·mL^(-1)时,对DPPH自由基和亚硝酸根离子清除率分别为80.3%和92.3%。响应面法优化的复合酶协同超声辅助提取青叶胆多酚工艺条件可靠,具有实际应用价值;青叶胆多酚有一定的抗氧化性,可作为一种抗氧化剂进行开发。 To determine the optimal extraction technology and antioxidant activity of Swertia mileens polyphenols(SMP)from green leaf cholesterol by enzyme-assisted ultrasound.Based on the data of Swertia mileens,a single factor test for designing a composite enzyme synergistic ultrasound-assisted extraction process was applied to screen the significant influencing factors,the Box-Benhnken center combination was used to design the response interview to determine the optimal process.The antioxidant activity of SMP was evaluated by determining its scavenging capacity on DPPH free radicals(DPPH·)and nitrite ions(NO2-).The results showed that the optimum conditions for extracting polyphenols from SMP by enzyme-assisted ultrasonography were enzymatic temperature 40℃,enzymatic time 50 min,liquid-to-material ratio 35∶1 mL·g^(-1),volume fraction of ethanol 63%,power 300 W,and amount of complex enzyme 6%(5%papain and 1%cellulase,based on the quality of green leaf biliary dry powder,the same below).Under these conditions,the extraction quantity of SMP was 17.50 mg·g^(-1).SMP had strong scavenging ability to DPPH free radicals and nitrite ions,and the scavenging ability increases with the increase of polyphenols mass concentration.The removal rates of DPPH free radicals and nitrite ions were 80.3%and 92.3%respectively at 0.007 mg·mL^(-1)of SMP.Response surface methodology the optimized composite enzyme synergistically ultrasonication assisted extraction of polyphenols from Swertia mileens reliable process conditions has practical applications,SMP has certain antioxidant activity and can be developed as an antioxidant.
