摘要
采用响应面法优化地参多糖的活性炭脱色工艺,并分析脱色后多糖的抗氧化性。以脱色率和多糖保留率为指标,采用综合评分法,在单因素试验基础上,利用Box-Benhnken中心组合设计和响应面分析法,系统考察了活性炭用量、脱色温度和脱色时间等因素对脱色效果的影响,建立了地参多糖脱色率的二次多项数学模型,并通过测定还原力来评价脱色后多糖的抗氧化性。结果表明,3个因素对地参多糖脱色效果的影响排序为脱色温度>脱色时间>活性炭用量;其优化脱色工艺为活性炭用量3.0%(以地参干粉质量为基准)、脱色温度25℃、脱色时间20 min。此条件下,多糖脱色率为57.42%,多糖保留率为78.35%;地参多糖具有一定的还原能力,其抗氧化活性弱于相同质量浓度的维生素C。
The activated carbon decolorization process of Lycopuslucidus Turcz. polysaccharides was optimized by response surface methodology,and the antioxidant activity of polysaccharides after decolorization was analyzed. Taking decolorization rate and polysaccharide retention rate as indicators,the comprehensive scoring method was adopted. On the basis of single factor test,the effects of the amount of activated carbon,decolorization temperature and decolorization time on decolorization were investigated by box Box-Benhnken central composite design and response surface analysis,and the second multiple mathematical model of Lycopuslucidus Turcz. Polysaccharide decolorization rate was established. The antioxidant activity of decolorized polysaccharide was evaluated by measuring its reducing power. The results showed that the influence of three factors on the decolorization of the polysaccharides was in the order ofdecolorization temperature > decolorization time > activated carbon dosage. The optimal decolorization process was as follows: the dosage of activated carbon 3. 0 %( with the quality of Lycopuslucidus Turcz.dry powder as benchmark),decolorization temperature 25 ℃,decolorization time 20 min. Under these conditions,the decolorization rate and retention rate of polysaccharides were 57. 42 % and 78. 35 % respectively. The Lycopuslucidus Turcz. polysaccharides had certain reducing ability and its antioxidant activity was weaker than that of vitamin C in the same concentration.
