摘要
目的:探讨当归多肽对慢性疲劳综合征小鼠抗疲劳的作用。方法:48只BALB/c小鼠随机分为空白对照组(8只)和慢性疲劳综合征模型组(40只)。慢性疲劳综合征模型组小鼠采取慢性束缚和冷水游泳,隔天交替进行,建立慢性疲劳综合征模型。造模成功后按随机数字表法分为模型对照组、当归多肽低剂量组、当归多肽中低剂量组、当归多肽高剂量组、红景天提取物组,每组8只。各给药组小鼠灌胃给予相应药物,空白对照组和模型对照组小鼠灌胃给予等体积的生理盐水,1次/d,连续给药4周。比较各组小鼠负重力竭游泳时间、甘油三酯(TG)、血尿素氮(BUN)、超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸(LD)、谷胱甘肽过氧化物酶(GSH-Px)、肝糖原储存量、腓肠肌糖原储存量、淋巴细胞增殖活性水平,核呼吸因子1(NRF-1)、线粒体转录因子A(TFAM)的mRNA表达水平,以及线粒体DNA(mtDNA)含量。结果:与模型对照组比较,红景天提取物组和当归多肽高、中、低剂量组小鼠负重力竭游泳时间均明显延长(P<0.05),SOD、GSH-Px、肝脏和腓肠肌糖原储存量均明显升高(P<0.05);与模型对照组比较,当归多肽高、中剂量组小鼠肝脏、肾脏、脾脏和胸腺的脏器指数及B淋巴细胞和T淋巴细胞活性均明显升高(P<0.05);与模型对照组比较,红景天提取物组和当归多肽中、高剂量组小鼠骨骼肌NRF-1 mRNA、TFAM mRNA相对表达量,线粒体DNA相对含量及GSH-Px、SOD活性均明显升高(P<0.05),血清TG、BUN、MDA、LD水平均明显降低(P<0.05);与红景天提取物组比较,当归多肽高剂量组小鼠负重力竭游泳时间明显延长(P<0.05),肝脏、心脏、肾脏、脾脏、胸腺指数,肝脏、腓肠肌糖原储存量,以及B淋巴细胞、T淋巴细胞活性均明显升高(P<0.05)。结论:当归多肽可以显著改善慢性疲劳综合征小鼠疲劳症状,且其效果优于红景天提取物。
Objective:To explore the anti-fatigue effect of Angelica sinensis polypeptide(ASP)on rats with chronic fatigue syndrome.Methods:A total of 48 BALB/c were randomly divided into blank control group(n=8)and chronic fatigue syndrome model group(n=40).Chronic restraint method and cold-water swimming method were used to establish the model of chronic fatigue syndrome alternately every other day.The chronic fatigue syndrome mice were randomly divided into model group,low-dose ASP group,medium-dose ASP group,high-dose ASP group and Rhodiola Rosea extract group,with 8 mice in each group.Mice in each treatment group were given corresponding drugs by gavage,and mice in blank control group and model group were given equal volume of normal saline by gavage,once a day,for 4 weeks.The weight bearing exhaustive swimming time,triglyceride(TG),blood urea nitrogen(BUN),superoxide dismutase(SOD),malondialdehyde(MDA),lactate(LD),glutathione peroxidase(GSH-Px),glycogen,sural muscle glycogen,lymphocyte proliferation activity,the mRNA expression of nuclear respiratory factor 1(NRF-1)and mitochondrial transcription factor A(TFAM),and the mitochondrial DNA(mtDNA)copy number of mice were measured.Results:Compared with the model group,the weight bearing exhaustive swimming time was prolonged(P<0.05),and the levels of serum SOD,serum GSH-Px,liver glycogen and gastrocnemius muscle glycogen were significantly increased in Rhodiola Rosea extract group,low-dose ASP group,medium-dose ASP group and high-dose ASP group(P<0.05).Compared with the model group, the organ index of liver, kidney, spleen and thymus, as well as the activity of B and T lymphocytes were significantly increased in the high-dose ASP group and medium-dose ASP group (P<0.05). Compared with the model group, the expression of NRF-1 mRNA and TFAM mRNA, as well as the mtDNA copy number in skeletal muscles and the levels of serum SOD and GSH-Px were significantly increased in Rhodiola Rosea extract group, medium-dose ASP group and high-dose ASP group (P<0.05).
