摘要
目的使用高通量超高效液相色谱-串联质谱(ultra-performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)建立人血浆中各胆固醇酯分子的测量方法,并对患者血浆样本实现定量分析。方法使用UPLC-MS/MS,以基质效应、检测限、回收率、精确度、准确度等方法学评价指标对13种胆固醇酯标准品进行方法学验证,同时收集来自首都医科大学附属北京安贞医院的稳定型冠心病患者、急性心肌梗死患者与体检中心对照人群血样各50例,检测血浆中13种胆固醇酯的分子水平,比较各组结果差异。结果通过对胆固醇酯标准品的方法学验证,本检测方案达到定量检测方法学验证要求。对人血样检测结果显示,胆固醇酯CE 22∶0、CE 22∶5、CE 20∶5、CE 18∶0、CE 18∶3等分子与冠心病的发生发展相关。结论基于UPLC-MS/MS高通量、高灵敏度和高特异性的检测特性,本文建立的胆固醇酯分子检测方法能够对血浆中多种胆固醇酯分子实现良好的分离和检测,为精准测量不同胆固醇酯分子提供了方法学参考。
Objective To use high throughput ultra-performance liquid chromatography-tandem mass spectrometry(UPLC/MS/MS)to establish and quantify the cholesteryl ester species in human plasma.Methods We used UPLC-MS/MS to verify the method’s detection and quantification limitation,matrix effect,recovery rate,accuracy and precision.We collected serum from the patients with stable coronary heart disease or acute myocardial infarction,and from health screening center visitors as control group.Plasma cholesteryl ester levels were measured and compared.Results The method was successfully validated and tested for plasma cholesteryl esters determination.Plasma cholesterol esters CE 22∶0,CE 22∶5,CE 20∶5,CE 18∶0 and CE 18∶3 were associated with disease occurrence and development of coronary heart disease.Conclusions By using a UPLC-MS/MS approach of high throughput,sensitivity and specificity,we set up a method for fine separation and detection of plasma cholesteryl esters,which potentially provides a quantification protocol for accurate measurement of plasma cholesteryl ester species for clinical.
