摘要
目的观察茯苓不同部位水提物对脾虚模型小鼠的健脾作用。方法将72只小鼠随机分为对照组,模型组,白茯苓低、高剂量组,赤茯苓低、高剂量组,茯苓皮低、高剂量组,阳性药组,每组8只。采用番泻叶水煎液加饥饱失常的复合因素法复制脾虚模型。比较各组小鼠体质量及肛温并对小鼠一般行为学进行观察,观察其脏器指数、粪便含水率、小肠推进率、外周血白细胞计数、血清D-木糖含量及白细胞介素-2(interleukin-2,IL-2)、肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)、分泌型免疫球蛋白(secretory immunoglobulin A,sIgA)水平的变化,并测定其结肠组织中密封蛋白(Claudin)、闭合蛋白(Occludin)的表达水平。结果与对照组比较,模型组小鼠体质量下降、粪便含水量上升(P<0.05);小鼠的脏器指数、D-木糖、sIgA含量及结肠组织中Claudin、Occludin蛋白表达水平均显著降低(P<0.05);小鼠小肠推进率、外周血白细胞计数及IL-2、TNF-α水平显著上升(P<0.05)。与模型组比较,各给药组上述指标均发生显著变化(P<0.05)。结论白茯苓可能通过调节免疫功能发挥健脾作用,而赤茯苓、茯苓皮通过调节机体的运化功能发挥健脾作用。
Objective To investigate the spleen-strengthening effect of water extracts from different parts of Poria cocos in a mouse model of spleen deficiency.Methods A total of 72 mice were randomly divided into control group,model group,low-and high-dose white Poria cocos groups,low-and high-dose Rubra Poria groups,low-and high-dose Cortex Poriae Cocos groups,and positive drug group,with 8 mice in each group.A model of spleen deficiency was established by the compound factor method with senna water decoction and irregular diet.Body weight and rectal temperature were compared between groups,and general behavioral observation was performed.Changes in related indices were measured,including visceral index,fecal water content,small intestine propulsion rate,peripheral blood leukocyte count,and the serum levels of interleukin-2(IL-2),tumor necrosis factor-α(TNF-α),and secretory immunoglobulin A(sIgA),and the expression levels of claudin and occludin in colon tissue were measured.Results Compared with the control group,the model group had a significant reduction in body weight and a significant increase in fecal water content,as well as significant reductions in visceral index,D-Xylose,sIgA content,and the protein expression levels of claudin and occludin in colon tissue(P<0.05)and significant increases in small intestine propulsion rate,peripheral blood leukocyte count,and the levels of IL-2 and TNF-α(P<0.05).Compared with the model group,the administration groups had significant changes in the above indices(P<0.05).Conclusion White Poria cocos may exert a spleen-strengthening effect by regulating immune function,while Rubra Poria and Cortex Poriae Cocos may exert such effect by regulating the body s transportation and transformation.
