黑曲霉发酵蚕沙产果胶酶工艺条件优化 被引量:2

Optimization of fermentation conditions for pectinase production from silkworm excrement by Aspergillus niger

摘要 蚕沙是蚕桑产业的主要废弃物,含有丰富的营养物质,是微生物发酵的良好原料。果胶酶是世界四大酶制剂之一,在工业生产中发挥着重要的作用。为了减少蚕沙随意丢弃带来的环境污染,充分挖掘蚕沙的利用价值,该研究以蚕沙为原料,利用黑曲霉发酵生产果胶酶。通过蚕沙固态发酵,首先采用单因素实验找出对果胶酶生产影响较大的因素,然后在单因素实验基础上利用正交实验对黑曲霉产果胶酶的条件进行优化。经单因素实验和正交实验得出黑曲霉固态发酵蚕沙的最佳产酶条件为:发酵初始pH 6,接种量0.5 mL,料液比(W/V)1∶1.7,基料(麸皮∶蚕沙)4∶6,发酵时间3 d,发酵温度30℃。在此发酵条件下,果胶酶活力达15.08 U/mL,是未优化前的1.87倍。研究结果可为桑蚕产业废弃资源的综合利用提供理论研究基础及新的思路。 Silkworm excrement is the main waste of sericulture industry. Silkworm excrement is a good raw material for microbial fermentation because it is rich in nutrients. Pectinase is one of the four major enzyme preparations in the world and plays an important role in industrial production. In order to reduce the environmental pollution caused by silkworm excrement discard and exploit the utilization of silkworm excrement,it was used as a raw material to produce pectinase by Aspergillus niger fermentation. With solid fermentation of silkworm excrement,single factor experiment was used to find out the factors that had great influence on pectinase production. The conditions of pectinase production by Aspergillus Niger were optimized by orthogonal experiment on the basis of single factor experiment. The optimal fermentation process conditions of solid fermentation of silkworm excrement by Aspergillus Niger were obtained as follows :initial fermentation pH 6,inoculum amount of Aspergillus niger 0.5 mL,ratio of medium material to water 1∶1.7(W/V),substrate ratio(wheat bran to silkworm excrement)4∶6,fermentation time 3 d and fermentation temperature 30 ℃. Under the optimized conditions,the enzyme activity reached 15.08 U/mL,which was 1.87 times higher than that before optimization. The results can provide theoretical basis and new ways for comprehensive utilization of waste resources in sericulture industry.
