摘要
从富含果胶质的果园的土壤中,通过对初筛的定性研究,对几种筛选培养基的比对选择和对复筛时Dp/D c值的测定,确定出一系列有效可行的筛选方法,从而获得一株能较强分解果胶质能力的菌株,然后对该菌株进行了初步的形态观察和鉴定。通过对发酵周期,温度,pH值几种因素对产酶影响的研究,经过对粗酶液的提取和对酶活的测定,确定了该菌株的基本发酵条件。在此条件下产酶最高可达到13968U/mL,从而最终确定了该菌株为一高产菌株。
From the soil of the high pectin, a strain which has high ability to break up pectin was selected. The works including initial selecting, the contrast and the choose of several selecting media and the measure of the Dp/Dc in the second selecting an effective and viable selecting method were done. The elementary morphology ob- servation and the identify of the strain were obtained. The effects of time, temperatue and pH on enzyme activity were studied. The yield of enzyme activity was 13968U/ml by liquid fermentation.
