摘要
异麦芽酮糖是一种多功能的新型甜味剂,同时也是国际上公认安全的蔗糖替代品,在医药、食品等行业中具有广阔的应用前景。蔗糖异构酶(sucrose isomerase,SIase)是生物转化生产异麦芽酮糖最有效的生物酶制剂。作者简要论述了异麦芽酮糖的生理特性及其在生产中现存的问题,详细阐述了SIase的来源、三维结构、催化机制、高效酶分子理性改造及其异源高效表达等。随后介绍了细胞及酶固定化在异麦芽酮糖生产中的广泛应用,并展望了蔗糖异构酶在生产异麦芽酮糖中的应用。
Isomaltulose is a new multifunctional sweetener and an internationally recognized safe sucrose substitute. Isomaltulose has wide application prospects in the medicine and food industries.Sucrose isomerase(SIase) is the most effective biological enzyme reagent for the production of isomaltulose by biological transformation. This review briefly presents the physiological characteristics of isomaltulose as well as the existing problems in its production. The source,three-dimensional structure, catalysis mechanism, rational design of high efficient enzymes and heterologous efficient expression of SIase were discussed in detail. In addition, the wide application of cell and enzyme immobilization in the production of isomaltulose was then introduced. Finally,the application of SIase in isomaltulose production was prospected.
