瑞香狼毒根部提取物对人肺腺癌A549细胞增殖及凋亡作用的影响

Effects of root extract of Stellera chamaejasme L.on the proliferation and apoptosis of human lung adenocarcinoma A549 cells

摘要 目的:探讨瑞香狼毒根部提取物(SCE)对人肺腺癌A549细胞作用和潜在机制。方法:细胞划痕和MTT用以评价和筛选SCE对A549细胞抑制的最佳浓度;激光共聚焦和荧光显微镜分别观察不同浓度SCE作用于A549后线粒体膜电位和染色质变化;Western blot检测p53、MDM2蛋白表达。结果:SCE药物组A549细胞较空白对照组在24、48 h迁移率均显著降低(P<0.01);在1~20μg/mL范围内,随着药物浓度的增加,A549细胞抑制率增强;与空白对照组比较,SCE低、中、高剂量组绿色荧光细胞占比逐渐增加;随着SCE浓度升高,A549细胞荧光增强至白色,染色质凋亡呈碎块状;与空白对照组比较,SCE低、中、高浓度组p53蛋白表达显著上升(P<0.05);与空白对照组和SCE低浓度组比较,SCE中、高浓度组MDM2蛋白表达显著下降(P<0.01)。结论:SCE能够显著抑制肺腺癌A549细胞增殖,其作用机制可能与作用于线粒体膜电位和p53/MDM2负反馈通路有关。 Objective:To investigate the effect and potential mechanism of root extract of Stellera chamaejasme L.(SCE)on the human lung adenocarcinoma A549 cells.Methods:Cell scratch and MTT were used to evaluate and screen the best concentration of SCE inhibiting A549 cells;The changes of mitochondrial membrane potential and chromatin were observed by laser confocal microscopy and fluorescence microscopy after SCE treatment with different concentrations on A549 cells;Western blot was used to detect the expression of p53 and MDM2 proteins.Results:The migration rate of A549 cells in SCE groups were significantly lower than that in blank control group at 24 h and 48 h(P<0.01);In the range of 1-20μg/mL,the inhibition rate of A549 cells increased with the increase of drug concentration;Compared with the blank control group,the proportion of green fluorescent cells in low,middle and high dose groups of SCE increased gradually;With the increase of SCE concentration,the fluorescence of A549 cells turned to white,and chromatin apoptosis became fragmented;Compared with the blank control group,the expression of p53 protein in low,medium and high dose groups of SCE increased significantly(P<0.05);Compared with the blank control group and low dose groups of SCE,the expression of MDM2 protein in medium and high dose groups of SCE decreased significantly(P<0.01).Conclusion:SCE could significantly inhibit the proliferation of lung adenocarcinoma A549 cells,and the mechanism may be related to effect on mitochondrial membrane potential and p53/MDM2 negative feedback pathway.
