摘要
目的:探讨肺康颗粒通过MST/hippo信号通路对慢性阻塞性肺疾病(COPD)大鼠炎症反应的影响及MST1/2在COPD大鼠中的表达状态。方法:通过持续烟熏建立大鼠COPD模型,设空白组,模型组,肺康颗粒低、中、高剂量组,XMU-MP-1组,地塞米松组。检测大鼠肺功能,支气管肺泡灌洗液中炎症因子CCL2、CXCL1、IL18、IL33、IL10水平以及肺组织中TLR4、MST1/2、IκB、p-IκB、p-Racl、Racl蛋白表达水平。结果:与模型组比较,肺康颗粒各剂量组肺功能显著改善(P<0.01,P<0.05),CCL2、CXCL1、IL18、IL33显著下降(P<0.01),IL10显著升高(P<0.01),MST1/2蛋白表达显著增加(P<0.05,P<0.01)。结论:COPD大鼠MST1/2表达下降,肺康颗粒可以上调COPD大鼠MST1/2表达,并可通过MST/hippo信号通路调控炎症。
Objective:To investigate the effects of Feikang Granules on inflammatory response of chronic obstructive pulmonary disease(COPD)rats through MST/hippo signaling pathway,and the expression status of MST1/2 in COPD rats.Methods:The COPD model of rats was established by continuous smoking.A blank group,a model group,a low-dose,mediumdose,high-dose Feikang Granules group,an XMU-MP-1 group,and a dexamethasone group were set up to detect the lung function of rats after the experiment,and the levels of inflammatory factors CCL2,CXCL1,IL18,IL33,and IL10 in bronchoalveolar lavage fluid,and the protein expression levels of TLR4,MST1/2,IκB,p-IκB,p-Rac1 and Rac1 in lung tissue.Results:Compared with model group,the lung function of Feikang Granules group was significantly improved(P<0.01,P<0.05),CCL2,CXCL1,IL18 and IL33 were significantly decreased(P<0.01),IL-10 was significantly increased(P<0.01),and the protein expression of MST1/2 was significantly increased(P<0.05,P<0.01).Conclusion:The expression of MST1/2 is decreased in COPD rats,and Feikang Granules can up-regulate the expression of MST1/2 in COPD rats,and regulate inflammation through MST/hippo signaling pathway.
