三七生药粉乙醇灭菌工艺优化及不同灭菌方式对质量的影响 被引量:4

Optimization of Ethanol Sterilization Technology for Notoginseng Radix et Rhizoma Powder and Impact of Sterilization Methods on Quality

摘要 目的:采用响应面法优选三七生药粉乙醇灭菌工艺,考察不同灭菌方式对三七生药粉灭菌效果及质量的影响。方法:以人参皂苷Rg1、人参皂苷Rb1及三七皂苷R1含量,皂苷总量和灭菌率的综合评分为指标,乙醇体积分数、乙醇用量、灭菌时间、干燥温度为因素,在单因素试验的基础上,采用响应面法优化三七乙醇灭菌工艺;分别对比湿热灭菌、辐照灭菌、干热灭菌和乙醇灭菌法对三七生药粉灭菌效果和质量的影响。结果:最佳灭菌工艺为乙醇体积分数74%、乙醇用量26%、灭菌时间2.2 h、干燥温度75℃;对比灭菌前后三七粉的质量,湿热灭菌和干热灭菌后三七中皂苷含量显著降低,其中湿热灭菌后皂苷含量已不符合《中华人民共和国药典》2020年版标准。指纹图谱显示,乙醇灭菌与生药材相似度较高,表明乙醇灭菌对三七影响不大。结论:乙醇灭菌法与其他灭菌方法相比,可在较低温度、更短时间、更加安全地得到符合规定的三七生药粉,且优选得到的灭菌工艺稳定可行、重复性好,可为其后续工艺开发和工业化生产提供参考。 Objective:To optimize ethanol sterilization technology for Notoginseng Radix et Rhizoma by response surface methodology and investigate the sterilization effect of different sterilization methods as well as their impact on the quality of Notoginseng Radix et Rhizoma powder.Methods:The ethanol sterilization technology for Notoginseng Radix et Rhizoma powder was optimized by response surface methodology based on the single factor test with the content of ginsenoside Rg1,ginsenoside Rb1,and notoginsenoside R1,the total saponins,and the comprehensive score of sterilization rate as the indicators,and the ethanol volume fraction,ethanol dosage,sterilization time,and drying temperature as factors.The sterilization effects of moist heat sterilization,irradiation sterilization,dry heat sterilization,and ethanol sterilization,as well as their impact on the quality of Notoginseng Radix et Rhizoma powder,were compared.Results:The optimal sterilization conditions were listed as follows:ethanol volume fraction of 74%,ethanol dosage of 26%,sterilization time of 2.2 h,and drying temperature of 75℃.As revealed by the quality of Notoginseng Radix et Rhizoma powder before and after sterilization,the content of saponins in Notoginseng Radix et Rhizoma decreased significantly after moist heat sterilization and dry heat sterilization,and the content after moist heat sterilization did not meet the standard in the Pharmacopoeia of the People's Republic of China(2020 edition).Fingerprints showed that ethanol sterilization could achieve similar results to that of crude medicinal materials,indicating that irradiation sterilization had little effect on Notoginseng Radix et Rhizoma.Conclusion:Compared with other sterilization methods,the ethanol sterilization method can obtain qualified Notoginseng Radix et Rhizoma powder at lower temperature and shorter time with higher safety,and the optimized sterilization technology is stable,feasible,and has good repeatability,which can provide a scientific basis for its subsequent process development
