马铃薯地方种质资源的SSR遗传多样性分析 被引量:8

SSR Analysis on Genetic Diversity for Local Potato Germplasm Resources

摘要 本研究利用SSR标记技术,对44份从湖北省收集的马铃薯地方种质资源进行遗传多样性分析,并构建其指纹图谱。研究结果显示,10对多态性丰富、条带清晰的引物,共扩增出122个标记位点,其中多态性位点有108个,多态性比率为88.5%。每对SSR引物扩增得到的多态性位点7~13个,平均10.8个。44份马铃薯地方种质资源间的遗传距离范围为0.0000~0.5085,平均遗传距离为0.2127。聚类结果显示,44份马铃薯地方种质资源在遗传相似系数为0.692时可被分为5大类,每大类分别包含9、21、11、2和1份材料。选用引物SKTIIKA和S192构建了马铃薯地方种质资源指纹图谱,可对马铃薯资源进行区分。本研究可为湖北省马铃薯种质资源的利用、育种亲本选配、品种鉴定和保护提供理论依据。 In this study,the genetic diversity analysis of 44 local potato germplasm resources collected from Hubei Province were conducted and the DNA fingerprinting were constructed by SSR markers.Ten SSR primers with rich polymorphism and clear bands were used for amplification of all experimental materials,and a total of 122 alleles were amplified,of which 108 were polymorphic sites,with the polymorphism ratio of 88.5%.The number of polymorphic sites amplified by each primer was 7~13,with an average of 10.8.The genetic distance of 44 local potato germplasm resources ranged from 0.0000 to 0.5085,and the average genetic distance was 0.2127.The clustering results showed that 44 local potato germplasm resources could be divided into 5 groups with the genetic similarity coefficient of 0.692,which contained 9,21,11,2 and 1 materials,respectively.The SSR primers SKTIIKA and S192 were used to construct DNA fingerprinting of local potato germplasm resources,which could distinguish potato resources.This study can provide a theoretical basis for the utilization of potato germplasm resources,breeding parent selection,variety identification and protection in Hubei Province.
