摘要
目的:探究IgA肾病(IgAN)血清长链非编码核糖核酸-lnc-TSI(lncRNA lnc-TSI)、长链非编码核糖核酸-生长停滞特异基因5(lncRNA GAS5)表达及与病情程度的相关性。方法:收集2019年1月-2021年1月四川绵阳四○四医院收治的125例IgAN患者的临床资料及血液样本,按照肾间质纤维化(RIF)程度不同分为轻度组(光镜下RIF面积<25%)、中度组(光镜下RIF面积25%~50%)、重度组(光镜下RIF面积>50%),另选35例体检健康人员血液样本作为对照组。比较各组血清lncRNA lnc-TSI、lncRNA GAS5水平,并分析其与IgAN患者肾功能及RIF程度关系。结果:IgAN患者各组24h尿蛋白、血肌酐(Scr)、尿素氮(BUN)均高于对照组,肾小球滤过率(eGFP)低于对照组,差异有统计学意义(P<0.05);且随RIF程度增加,24 h尿蛋白、Scr、BUN逐渐升高,eGFR逐渐降低,差异有统计学意义(P<0.05);随着RIF程度增加,血清lncRNA lnc-TSI、lncRNA GAS5表达水平逐渐降低,差异有统计学意义(P<0.05)。Spearman相关性分析显示,IgAN患者血清lncRNA lnc-TSI、lncRNA GAS5水平与RIF程度呈负相关,差异有统计学意义(P<0.05)。结论:血清lncRNA lnc-TSI、lncRNA GAS5表达水平在IgAN患者中较低,且与患者RIF程度具有一定相关性,能有效监测和评估患者病情进展。
Objective:To investigate the expression of serum long-chain non-coding ribonucleic acid-lnc-TSI(lncRNA lnc-TSI)and long-chain non-coding ribonucleic acid-growth arrest-specific gene 5(LncRNA GAS5)in IgA nephropathy(IgAN)and the correlation with the extent of the disease.Methods:Clinical data and blood samples were collected from 125 patients with IgAN admitted to Mianyang 404th Hospital from January 2019 to January 2021.According to the different degrees of renal interstitial fibrosis(RIF),they were divided into mild group(RIF area under light microscopy<25%),moderate group(RIF area under light microscopy 25%-50%),and severe group(RIF area under light microscopy>50%).Another 35 blood samples from healthy individuals with physical examination were selected as the control group.The serum lncRNA lnc-TSI and LncRNA GAS5 levels of each group were compared and analyzed in relation to the renal function and the degree of RIF in IgA patients.Results:The24 h urine protein,blood creatinine(Scr)and urea nitrogen(BUN)were higher in all groups of IgA nephropathy patients than in the control group,and the glomerular filtration rate(eGFP)was lower than that of the control group,with statistically significant differences(P<0.05).The expression levels of serum lncRNA lnc-TSI and LncRNA GAS5 decreased gradually with the increase of RIF degree.It showed that serum lncRNA lnc-TSI and LncRNA GAS5 levels in patients with IgA nephropathy were negatively correlated with the degree of RIF,and the difference was statistically significant(P<0.05).Conclusion:The expression levels of serum lncRNA lnc-TSI and LncRNA GAS5 were low in patients with IgA nephropathy and correlated with the degree of RIF,which can effectively monitor and assess the progress of patients’disease.
