一株短小芽胞的分离鉴定 及其对屠宰牛血液的降解功能研究

摘要 本试验旨在通过筛选高效降解牛血液的菌株,以生物方式降解血液中大分子蛋白物为多肽、氨基酸等小分子产物,再制备为含氨基酸水溶肥料,从而实现屠宰牛血液的无害化环境处理和废弃蛋白资源再利用。本研究以屠宰场血液污泥为分离基质,通过酪素培养基平板和哥伦比亚血琼脂平板筛选出1株能够高效降解血液蛋白的菌株,经形态学特征、生理生化和16S rDNA序列鉴定为短小芽胞(Bacillus pumilus NWMCC0302),该菌株的16S rDNA序列在GenBank中登录号为OL636364。通过单因素试验确定短小芽胞降解牛血液的关键因素,再采用Plackett-Burman试验和Box-Behnken试验设计优化其最佳降解工艺条件。结果表明:降解的关键影响因素为初始pH、麦麸浓度、温度和接种量。最佳降解工艺条件为初始pH 7.39,牛血液体积分数10%(V/V),麦麸浓度21.31 g/L,接种量2.05%(V/V),降解温度38.11℃,对牛血液蛋白的降解率为53.83%。试验筛选出的短小芽胞具有高效降解牛血液蛋白的能力,可以为屠宰废弃血液资源的生物方式处理和循环利用提供候选菌株。 This study screened strains that can degrade bovine blood with high efficiency to biodegrade macromolecular proteins in the blood into small molecular products(peptides and amino acids),which can be prepared for water-soluble fertilizers to realize the harmless environmental treatment of slaughtered bovine blood and reuse of waste protein resources.A strain capable of efficiently degrading blood protein was isolated from slaughterhouse blood sludge by a casein medium and Colombian blood agar plates and was identified as Bacillus pumilus NWMCC0302 via morphological,physiological,biochemical,and 16S rDNA sequence analysis(registered as OL636364 in the GenBank).A single factor experiment determined the critical factors of Bacillus pumilus degrading bovine blood,and the Plackett-Burman experiment and Box-Behnken experiment optimized the degradation process conditions.The results showed that the essential factors affecting degradation were initial pH,wheat bran concentration,inoculation amount and temperature.The optimum degradation conditions were initial pH 7.39,bovine blood volume fraction 10%(V/V),wheat bran concentration 21.31 g/L,inoculum 2.05%(V/V),and degradation temperature 38.11℃.Under the optimized conditions,the degradation rate of bovine blood protein could reach 53.83%.The Bacillus pumilus screened in this study could efficiently degrade bovine blood proteins,which could be used as a candidate strain for the biological treatment and recycling waste blood resources in slaughter.
