摘要
以芳樟一年生半木质化枝条为材料,修剪成一叶一芽茎段,研究3000 mg/L IBA处理下芳樟微扦插生根过程相关氧化酶活性、可溶性糖和淀粉含量的变化.结果表明:3000 mg/L IBA处理下芳樟微扦插生根达到67.67%,生根过程分为愈伤组织形成期、生根诱导期和表达期3个阶段.芳樟微扦插生根和SOD、POD、PPO、IAAO酶活性密切相关.扦插(0~21)d,SOD、POD、PPO活性升高,IAAO活性下降至最低值,可能有利于芳樟插穗愈伤组织的形成;扦插(21~28)d,SOD、POD活性继续上升至峰值,PPO活性开始下降,IAAO活性升高,有利于芳樟插穗基部愈伤组织增大和根原基诱导;扦插28 d后,SOD、POD、PPO活性缓慢下降后趋于稳定,IAAO活性缓慢上升后趋于稳定,有利于芳樟插穗不定根的形成.可溶性糖和淀粉含量对插穗生根起着重要作用,扦插(0~14)d,淀粉含量迅速下降,之后淀粉含量逐渐升高;而可溶性糖在扦插(0~28)d迅速上升,之后基本保持不变.
The one-year-old semi-lignified branches of Cinnamomum camphora ct.linalool was pruned into stem segment with one leaf and one bud,in order to study the variation trend relevant oxidase activities and carbohydrates during the rooting process of micro-cuttage of Cinnamomum camphora ct.linalool with 3000 mg/L IBA treatment.The results show that the rooting rate of micro-cuttage of Cinnamomum camphora ct.linalool is 67.67%with 3000 mg/L IBA treatment,and the rooting process is divided into callus formation stage,rooting induction stage and expression stage.The rooting process of cuttings is closely related to the activities of SOD,POD,PPO and IAAO.0 to 21 d of cutting,the activities of SOD,POD and PPO increase,and the activity of IAAO decrease to the bottom,which is beneficial to the callus formation of Cinnamomum camphora ct.linalool cuttings.21 to 28 d of cutting,the activities of SOD and POD raise to the peak,the activities of PPO began to decrease,and the activities of IAAO increase,which is beneficial to the enlargement of callus and root primordium induction of Cinnamomum camphora ct.linalool cuttings.After 28 d of cutting,the activities of SOD,POD and PPO decrease slowly and then stabilize,while the activities of IAAO increase slowly and then stabilize,which is beneficial to the formation of adventitious roots.Soluble sugar and starch content playe an important role in rooting of Cinnamomum camphora ct.linalool cuttings.0 to 14 d of cutting,the starch content decrease and then increase gradually,while the content of soluble sugar increases rapidly from 0 to 28 d of cutting,and remains basically unchanged.
