摘要
以垂珠花(Styrax dasyanthus )1~2年生枝条为扦插材料,对其枝条上、中、下3部分插穗韧皮部内生根抑制物水浸提液进行化感活性的测定,并采用三因子正交试验研究了生长调节剂吲哚乙酸(IAA)、吲哚丁酸(IBA)、α-萘乙酸(NAA)和2,4-二氯苯氧乙酸(2,4-D)对垂珠花扦插生根及生根过程中吲哚乙酸氧化酶(IAAO)和多酚氧化酶(PPO)活性的影响.结果表明:垂珠花插穗上、中、下3部分韧皮部内存在某些物质在不同程度上抑制白菜籽萌发及其生长发育,且不同部位间的抑制程度亦呈现出显著差异.下部插穗中该物质的含量相对较高,故可能对插穗生根起到抑制作用,故垂珠花扦插繁殖材料宜选用枝条中上部为佳.初步判定,垂珠花插穗生根类型属于皮部生根型.4种生长调节剂处理组合均能显著提高垂珠花插穗的各生根指标,其中,500 mg/L IBA 处理30 min对垂珠花硬枝扦插生根率促进作用最为显著,插穗生根率最高,其生根率、生根数和最长根长分别为62.25%、7.53条和93.29 mm;其次是 IAA,NAA处理效果较差.经 IBA处理后可提高垂珠花插穗 PPO 活性,降低 IAAO 活性,有效缩短垂珠花插穗生根时间,显著提高其生根率.
By using the water solution from three parts of branches of phloem, the effects of extract from cuttings of Styrax dasyanthus, and the effects of plant growth regulators on the hardwood-cutting rooting of S. dasyanthus were studied with the orthogonal design of three factors. The dynamic changes of activities of indoleacetic acid oxidase (IAAO) and polyphenol oxidase (PPO) during rooting process were also investigated. The results showed that:Top, middle and basal parts of cutting existed some materials in the phloem, which inhibited seed germination and growth of Chinese cabbage in different extent, and the in- hibition among different parts of branches also had obviously differences, the contents of the materials rela- tively higher in the basal part cutting, and it influenced and inhibited rooting of cuttings, so the cuttings should be selected from middle to upper of the branch for propagation material. S. dasyanthus is the plant of the surface root type by preliminary judgment. Four kinds of growth regulators could significantly im- prove the cuttings rooting index. The most significant effect on hardwood cutting by the treatment of 500mg/L IBA 30min, the rooting rate was the highest, and rooting rate, root number and the longest root length were 62. 25 %,7.53 and 93.29 mm, respectively. IAA and NAA treatments showed less effects. The IBA treatment increased the PPO activity, but reduced the IAAO activity, so it could effectively reduce the cuttings rooting time and increase the rooting rate.
