摘要
目的基于网络药理学探讨芪参益气滴丸治疗糖尿病合并冠心病心绞痛的作用机制。方法采用中药系统药理学数据库分析平台(TCMSP)检索芪参益气滴丸的活性成分及靶点,在DisGeNET数据库检索“糖尿病”“冠心病”“心绞痛”靶点,对冠心病和心绞痛靶点合并去重后与糖尿病靶点取交集,获得疾病靶点。对药物成分与疾病的靶点取交集,得到芪参益气滴丸治疗糖尿病合并冠心病心绞痛的作用靶点。通过STRING平台获取蛋白基因互作数据,利用Cytoscape软件构建蛋白互作网络,采用DAVID数据库对作用靶点进行基因本体(GO)功能、京都基因和基因组百科全书(KEGG)通路富集分析。结果芪参益气滴丸有110个活性成分和255个作用靶点,其中,102个活性成分作用于106个靶点的20条信号通路治疗糖尿病合并冠心病心绞痛。生物功能和通路富集结果提示芪参益气滴丸可以调控RNA聚合酶Ⅱ启动子转录、凋亡过程、细胞增殖、炎症反应、缺氧反应、基因表达、血管新生等生物过程,作用于磷脂酰肌醇3-激酶(PI3K)-蛋白激酶B(Akt)、低氧诱导因子-1(HIF-1)、叉头状转录因子O(FoxO)、Toll样受体、腺苷酸活化蛋白激酶(MAPK)、核转录因子-κB(NF-κB)、Janus激酶(JAK)-信号转导及转录激活因子(STAT)、血管内皮生长因子(VEGF)、细胞凋亡、p53等多条信号通路。结论芪参益气滴丸可能通过促进血管再生、降低细胞凋亡、抑制炎症反应和减轻氧化应激改善糖尿病合并冠心病心绞痛症状。
Objective To explore the mechanism of Qishen Yiqi Droplet in the treatment of diabetes mellitus complicated with coronary heart disease angina pectoris based on network pharmacology.Methods The Traditional Chinese Medicines Systems Pharmacology Platform(TCMSP)was used to search for the active ingredients and targets of Qishen Yiqi Droplet,and the DisGeNET was used to search for targets of"diabetes","coronary heart disease",and"angina pectoris".After the coronary heart disease angina pectoris targets were combined and de-duplicated,they were intersected with diabetes targets to obtain disease targets.The target of Qishen Yiqi Droplet in the treatment of diabetes with coronary heart disease angina pectoris was obtained by intersection of the components of drug and the target of disease.The protein gene interaction data was obtained through STRING platform,the protein interaction network was constructed by Cytoscape software,and the gene ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis of action targets were conducted by DAVID.Results There were 110 active components and 255 targets in Qishen Yiqi Droplet,of which 102 active components act on 20 signaling pathways of 106 targets in the treatment of diabetes mellitus complicated with coronary heart disease angina pectoris.Biological function and pathway enrichment results suggested that Qishen Yiqi Droplet could regulate RNA polymeraseⅡpromoter transcription,apoptosis,cell proliferation,inflammation,hypoxia,gene expression,angiogenesis,and act on phosphatidylinositol 3-hydroxykinase(PI3K)-protein kinase(Akt),hypoxic inducible factor 1(HIF-1),forkhead box protein O(FoxO),Toll-like receptor,mitogen-activated protein kinase(MAPK),nuclear factor-κB(NF-κB),Janus kinase(JAK)-signal transducer and activator of transcription(STAT),vascular endothelial growth factor(VEGF),apoptosis,p53,and other signal pathways.Conclusion Qishen Yiqi Droplet may improve the symptoms of diabetes mellitus complicated with coronary heart disea
