摘要
为探讨不同时期藏绵羊瘤胃热休克蛋白70家族基因的表达特征及瘤胃微生物菌群结构在瘤胃免疫功能中的关系。采用Real-time PCR技术和16S rRNA测序对放牧藏绵羊不同时期(4月、6月、10月和12月)热休克蛋白70家族基因(HSPA1A和HSPA8)(n=12)的表达特征和瘤胃微生物菌群(n=24)进行分析,并采用Spearman对瘤胃微生物与基因表达进行相关性分析。结果表明,不同时期HSPA1A和HSPA8表达量存在显著差异,HSPA1A和HSPA8表达量在4月和6月显著高于10月和12月;不同时期的瘤胃微生物丰度和多样性存在显著差异,Bacteroidetes、Rikenellaceae_RC9_gut_group在12月显著高于6月和10月;Firmicutes、Succiniclasticum和Butyrivibrio_2在6月显著高于4月和12月。相关性分析发现,不同时期瘤胃微生物与基因表达存在一定相关性,即瘤胃微生物在一定程度上可以对宿主的瘤胃免疫功能产生影响,这为藏绵羊瘤胃微生物与宿主免疫的互作研究提供基础数据。
To explore the relationship between the expression characteristics ofrumen heat shock protein 70 family genes and the structure of rumen microflora in rumen immune function of Tibetan sheep in different periods,Real-time PCR technology and 16S rRNA sequencing were used to analyze the expression characteristics of heat shock protein 70 family genes(HSPA1A and HSPA8)(n=12)in different periods of grazing Tibetan sheep(April,June,October,and December).The correlation between rumen microbes and gene expression was analyzed by Spearman.The results showed that there were significant differences in the expression levels of HSPA1A and HSPA8 in different periods.The expression levels of HSPA1A and HSPA8 in April and June were significantly higher than that in October and December(P<0.05).There were significant differences in the abundance and diversity of rumen microbes in different periods.Bacteroidetes and Rikenellaceae_RC9_gut_group were significantly higher in December than that in June and October(P<0.05);Firmicutes,Succiniclasticum and Butyrivibrio_2 were significantly higher in June than that in April and December(P<0.05).The correlation analysis found that there was a certain correlation between rumen microbes and gene expression in different periods,that is,rumen microbes could affect the immune function of the host to a certain extent.This study provides the basic data for the interaction between rumen microbes and host immunity in Tibetan sheep.
