摘要
2017年5月中旬在河南省信阳市东南地区石山口水库两支流处分别设置1^(#)和2^(#)两个采样点,坝前设3^(#)采样点,采用高通量测序法研究了微生物多样性,分析了多样性指数,包括α多样性指数分析(ace指数、chao指数、shannon指数和simpson指数)和β多样性指数分析(主成分分析和聚类分析),分析出单个样本的微生物丰度和均匀度和多个样本间的微生物群落差异。结果表明,石山口水库1^(#)采样点的0 m处微生物群落组成与其他采样点差异较大。2^(#)采样点的0 m、3 m处归聚为一类,说明这两处采样点微生物群落组成较为相似。3^(#)采样点的0 m、4 m处归聚为一类,说明这两处微生物群落结构组成较为相似。1^(#)采样点的3 m处与3^(#)采样点的8 m处归聚为一类,微生物组成较为相似。总的来说,石山口水库水体中不同层次的微生物群落结构组成差异不显著,水深对微生物多样性影响较大,水体中微生物以念珠菌属(Candidatus-Pelagibacter)、硫单胞菌属(Thiomonas)、军团杆菌属(Legionella)等为优势种,而不同断面样品的优势菌属有所不同;石山口水库微生物群落丰度总体较高,多样性也相对较高,其中1^(#)采样点3 m处与3^(#)采样点8 m处的Shannon指数高于其他采样点,表明水体深度是影响其微生物群落结构的主要因子。本研究结果可对确定水质的优劣,以及石山口水库微生物多样性和渔业养殖模式研究提供数据支撑和理论指导。
In the middle of May 2017,two sampling sites 1^(#)and 2^(#)were set at two tributaries of Shishankou Reservoir in southeast area of Xinyang City,Henan Province,and 3^(#)were set in front of the dam.The microbial diversity was studied by high-throughput sequencing method and the diversity index was analyzed.The α-diversity index(ACE index,Chao index,Shannon index and Simpson index)and β-diversity index(principal component analysis and cluster analysis)were used to analyze the microbial abundance and evenness of a single sample and the microbial community differences among multiple samples..The results show that the sampling point 1 of Shishankou reservoir is very different from other sampling points.The sampling sites at 0 m and 3 m at No.2 can be grouped into one category,indicating that the microbial communities at these two sampling sites are relatively similar.The 0 m and 4 m locations of sampling site No.3 can be grouped into one category,indicating that the two microbial community structures are relatively similar.The sampling point 1 at 3 m and the sampling point 8 at 8 m can be grouped together,and the microbial composition is relatively similar.In general,the composition of microbial community structure at different levels in the water body of Shishankou reservoir is not very different.Water depth has a greater impact on microbial diversity.The microorganisms in the water body are Candidatus-Pelagibacter,Thiomamonas,Legionella and other strains as the dominant species,and the dominant bacteria of different cross-section samples are different.The abundance of microbial communities in Shishankou reservoir is generally high and the diversity is relatively high.Among them,the Shannon index at 3 m of sampling point 1 and 8 m of sampling point 3 are higher than other sampling points.The analysis shows that the depth of the water body is the main factor affecting its microbial community structure.The research results provide a series of data support and theoretical guidance for determining the quality of water
