冷应激家兔血压与血清细胞因子的变化及其相互关系 被引量:10

摘要 目的研究冷应激家兔血压和血清中白细胞介素-2(IL-2)、白细胞介素-10(IL-10)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的变化及其相互关系,为今后预防或降低冷应激引发的高血压及肿瘤等疾病提供理论依据。方法将清洁级中国本地白家兔麻醉后,测定冷应激[(3.0±1.0)℃]0、0.25、0.50、0.75、1.00、1.50、2.00、3.00、4.00和6.00h家兔的平均动脉血压(mean artery pressure,mAP)、平均收缩压(mean systolic pressure,mSP)和平均舒张压(mean diastolic pressure,mDP),以及血清中IL-2、IL-10、IFN-γ、TNF-α的含量。结果随着冷应激时间的延长,mAP、mSP和mDP呈波动性变化,整体呈下降的趋势。在冷应激1h时,上升到最大值,但仍低于对照组。冷应激家兔血中IL-2的浓度呈先升高后降低的趋势;IL-10的浓度呈先降低后升高的趋势;TNF-α、IFN-γ的浓度呈下降的趋势。冷应激家兔血清中IFN-γ与TNF-α间呈正相关(r=0.9830,P<0.0001)。以IFN-γ为自变量(x),TNF-α为应变量(y),建立线性回归模型为:y=2.37+0.18x(R2=0.9964)。冷应激家兔血压(mAP、mSP、mDP)与血清中IL-2之间呈正相关(均0.2<r<0.3);与血清中IL-10之间呈负相关(均0.7<r<0.8);与血清中TNF-α、IFN-γ之间均呈正相关(均0.5<r<0.6)。结论在冷应激状态下,家兔机体的生理机能和免疫功能均发生一定程度的变化,从而使其最大程度地减少应激性损伤,冷应激家兔血压与血清中IL-2、IL-10、IFN-γ及TNF-α之间存在不同程度的相关关系,其机制有待进一步研究。 Objective To know the effect of cold stress on the artery pressure and the concentration of IL-2, IL-10, IFN-γ, TNF-α in the serum of the rabbits. Methods The local white rabbits of China (clean grade) were anesthetized. The mAP (mean artery pressure), mSP (mean systolic pressure), mDP (mean diastolic pressure) and the concentration of IL-2, IL-10, IFN-γ, TNF-α were determined at 0 h, 0.25 h, 0.50 h, 0.75 h, 1.00 h, 1.50 h, 2.00 h, 3.00 h, 4.00 h and 6.00 h after cold stress. Results The mAP,mSP and mDP changed undulant with the delay of cold stress,but the whole tendency was descendent. The mAP,mSP and mDP went up to the peak at 1 h,however lower than control. The concentration of IL-2 increased then decreased with the delay of cold stress,the concentration of IL-10 went down then up and the concentration of TNF-α,IFN-γ went down gradually. There was a strong correlation between IFN-γ and TNF-α (r=0.983 0,P<0.000 1). The linear regression model between the concentration of IFN-γ and TNF-α was y=2.37+0.18x (IFN-γ was the independent variable and TNF-α was the dependent variable,R2=0.996 4),There was a weak correlation between the blood pressure (mAP,mSP,mDP) and IL-2 (0.2<r<0.3); There was strong negative correlation between the blood pressure (mAP,mSP,mDP) and IL-10 (0.7<|r|<0.8). There was moderate correlation between the blood pressure (mAP,mSP,mDP) and IFN-γ,TNF-α (0.5<r<0.6). Conclusion The physiological and immunological function of the rabbit changed during the cold stress so that the damage induced by cold stress should be decreased. There is a certain correlation among the blood pressure (mAP,mSP,mDP) and IL-2,IL-10,IFN-γ,TNF-α in serum of the rabbits. The related mechanisms should be studied in the future.
