摘要 目的:建立西洋参标准汤剂质量评价方法,为其质量控制提供参考。方法:采用传统煎煮方法,制备15批西洋参标准汤剂,以人参皂苷Rg_(1)、Re、Rb_(1)、Rc、Rb_(2)、Rd含量、转移率、出膏率及HPLC指纹图谱相似度为评价指标,建立质量评价体系。采用主成分分析法和灰色关联度法对指纹图谱进行分析。结果:15批西洋参标准汤剂中人参皂苷Rg_(1)、Re、Rb_(1)、Rc、Rb_(2)、Rd的含量分别为0.47~1.46 mg/g、4.42~6.52 mg/g、5.59~10.19 mg/g、0.91~2.07 mg/g、0.41~0.88 mg/g、0.57~1.74 mg/g;出膏率为31.09%~40.13%;6个成分的转移率范围分别为42.34%~89.03%、41.89%~81.42%、48.30%~103.35%、43.41%~92.41%、21.93%~47.83%、44.74%~77.83%;指纹图谱相似度均≥0.920,确定15个共有峰,指认了6个成分;主成分分析法提取出2个主成分,其累计方差贡献率为90.119%,可代表样品的大部分信息。指纹图谱中6个成分的灰色关联度为0.582~0.937,加权灰色关联度为0.445~0.930。排序结果显示S14号样品质量最佳。结论:该研究建立的质量评价方法稳定、可靠,能有效地对西洋参标准汤剂进行综合质量评价,为西洋参标准汤剂及其制剂的质量控制提供了依据。 Objective:To establish the quality evaluation method of Panacis Quinquefolii Radix standard decoction,so as to provide reference for its quality control.Methods:Fifteen batches of Panacis Quinquefolii Radix standard decoction were prepared by traditional decocting method.The quality evaluation system was established with the contents of ginsenoside Rg_(1),Re,Rb_(1),Rc,Rb_(2)and Rd,transfer rate,extraction rate and similarity of HPLC fingerprint as evaluation indexes.Principal component analysis and grey relational analysis were used to analyze the fingerprint.Results:The contents of ginsenoside Rg_(1),Re,Rb_(1),Rc,Rb_(2)and Rd in 15 batches of Panacis Quinquefolii Radix standard decoction were 0.47~1.46 mg/g,4.42~6.52 mg/g,5.59~10.19 mg/g,0.91~2.07 mg/g,0.41~0.88 mg/g,0.57~1.74 mg/g,respectively.The extract rate ranged from 31.09%to 40.13%.The transfer rates of the six components were in the range of 42.34%~89.03%,41.89%~81.42%,48.30%~103.35%,43.41%~92.41%,21.93%~47.83%,44.74%~77.83%,respectively.The similarity of fingerprints was greater than or equal to 0.920.Fifteen common peaks were identified and six of them were identified.Two principal components were extracted by principal component analysis,the cumulative variance contribution rate was 90.119%,which could represent most of the information of the samples.The grey correlation degree of the six components in the fingerprint was 0.582~0.937,the weighted grey correlation degree was 0.445~0.930.The sequencing results showed that sample S14 had the best quality.Conclusion:The quality evaluation method established in this study is stable and reliable,which can effectively evaluate the comprehensive quality of Panacis Quinquefolii Radix standard decoction,and provide a scientific basis for the quality control of Panacis Quinquefolii Radix standard decoction and its preparations.
