基于标准汤剂指纹图谱分析的糖肾清毒颗粒质量评价 被引量:6

摘要 目的:建立糖肾清毒颗粒标准汤剂、中间体、成品的HPLC指纹图谱,研究三者的相关性并进行多成分定量分析。方法:色谱柱为Inert Sustain C18柱(250 mm×4.6 mm,5μm),采用多波长切换技术,以乙腈-0.1%磷酸溶液为流动相进行梯度洗脱,流速1.0 m L/min,柱温35℃。采用"中药色谱指纹图谱相似度评价系统软件(2012版)"分别对10批标准汤剂、中间体及成品的指纹图谱进行相似度评价,并将对照图谱与单味药材进行谱峰匹配,确定共有峰归属,通过混合对照品对共有峰进行成分指认并对其进行定量分析。结果:10批标准汤剂、中间体及成品的指纹图谱与各自对照图谱的相似度均大于0.95,标准汤剂标定有21个共有峰,中间体、成品标定有20个共有峰,所有共有峰都能归属到药材。标准汤剂共有峰中,8个来源于黄芩,1个来源于牛蒡子,9个来源于川银花,3个来源于蒲公英,2个来源于豨莶草。通过对照品指认了7个共有峰,分别为绿原酸(3号峰)、咖啡酸(5号峰)、黄芩苷(14号峰)、牛蒡苷(15号峰)、汉黄芩苷(19号峰)、黄芩素(20号峰)和汉黄芩素(21号峰)。标准汤剂、中间体及成品的指纹图谱与对照图谱的相似度较高,相关性良好。不同批次的指标成分含量变化小,成品质量稳定。结论:建立的指纹图谱相似度评价结合多指标成分定量分析的方法可为糖肾清毒颗粒的质量控制与鉴别提供依据。 Objective :To establish an HPLC fingerprint and analyze the correlation of standard decoction,intermediate,and finished products of Tangshen Qingdu granular,to determine the contents of the multi-index components for its quality control.Methods: The separation was performed on Inert Sustain C18 column(250mm×4.6mm,5μm)with gradient elution using acetonitrile-0.1% phosphoric acid as the mobile phase and adopting multi-wavelength switch technology.The flow rate was 1.0mL/min,the column temperature was 35%.The Similarity Evaluation System for Chromatographic Fingerprint of TCM(2012edition)was used to analyze the similarity degree of standard decoction,intermediate,and finished products,the peak pattern matching with the original medicine was conducted.The common peaks were identified by reference compounds and the content was detected.Results :The fingerprints of ten batches of standard decoction,intermediate,and finished products were established,and the similarities to the common mode were all over 0.95.Totally 21 common peaks were identified in the standard decoction,20 common peaks were identified in intermediate and finished products,and all common peaks belonged to herbs.In standard decoction,eight mutual peaks were stem from Scutellariae Radix,one mutual peak was stem from Arctii Fructus,nine mutual peaks were stem from Lonicera similis,three mutual peaks were stem from Taraxaci Herba,and two mutual peaks were stem from Siegesbeckia Herba.Based on the retention time of reference substances,seven constituents including chlorogenic acid (peak 3),caffeic acid (peak 5),baicalin (peak 14),arctiin (peak 15),wogonoside (peak 19),baicalein(peak 20)and wogonin (peak 21)were identified.The similarity between the reference fingerprint and fingerprints of standard decoction,intermediate,and finished products was high,presenting a good correlation.The contents of the indexes of different batches was small and the quality of the finished products was stable.Conclusion:Similarity evaluation of fingerprint combined with mul
