紫菀水提液对肺炎支原体感染小鼠肺组织病理变化、细胞凋亡及TGF-β1表达的影响 被引量:7

摘要 目的:通过研究紫菀水提液对肺炎支原体感染小鼠肺组织病理变化、细胞凋亡及转化生长因子TGF-β1表达的调控作用,从而阐明其对肺炎支原体肺炎的作用机制。方法:C57小鼠60只,随机分为6组,分别是正常对照组、模型对照组、阳性对照组(阿奇霉素组)和紫菀水提液高、中、低剂量组,每组10只。采用滴鼻法制备小鼠肺炎支原体肺炎模型,紫菀水提液低、中、高剂量组分别予0.49、0.98、1.95 g/kg灌胃,阿奇霉素组灌胃给药剂量为32 mg/kg,每天1次,连续给药14 d,正常对照组及模型对照组小鼠灌胃等量生理盐水。HE染色观察小鼠肺组织病理改变,流式细胞仪检测各组小鼠肺组织细胞凋亡率,Real-time PCR实验检测小鼠肺组织TGF-β1mRNA表达。结果:HE染色结果显示,与模型对照组比较,各给药组小鼠肺组织病理损伤明显减轻;流式细胞凋亡实验结果表明,各给药组小鼠肺组织细胞凋亡率显著低于模型对照组;Real-time PCR结果显示,各给药组小鼠肺组织的TGF-β1 mRNA表达量明显低于模型对照组。结论:紫菀水提液能够减轻肺炎支原体肺炎对小鼠肺组织造成的病理损伤,降低肺组织细胞凋亡率和肺组织转化生长因子TGF-β1表达是其部分作用机制。 Objective:To explore the mechanism of Aster aqueous extract on Mycoplasma pneumoniae pneumonia by studying the regulatory effects of Aster extract on the pathological changes,apoptosis and expression of the transforming growth factor TGF-β1 in mice infected with Mycoplasma pneumoniae.Methods:60 C57 mice were randomly divided into 6 groups of 10 mice,namely normal control group,model control group,Aster high,medium and low dose group,and posotive group(Azithromycin).The models were coped by nasal drip Mycoplasma pneumoniae(1×106CCU),and the low,medium and high doses Aster groups were given Aster aqueous extract 0.49,0.98,1.95 g/kg,respectively,the positive group was given Azithromycin 32 mg/kg and continuous administration for 14 days.Mice in the normal control and the model control group were given an equal amount of normal saline per day.The pathological changes of lung tissue in each group mice were observed by HE staining,the apoptosis rate of lung tissue cells was detected by the flow cytometry and the expression of TGF-β1 mRNA in mice’s lung tissue was detected by Real-time PCR.Results:The HE staining results showed that compared with the model control group,the pathological damage of the lung tissue of mice in each medication group was significantly reduced,The results of flow cytometry showed that the apoptosis rate of lung tissue cells of mice in each medication group was significantly lower than that of model control group,Real-time PCR results showed that TGF-β1mRNA expression in lung tissue of mice in each medication group was significantly lower than that of the model control group.Conclusion:Aster aqueous extract can alleviate the pathological damage caused by Mycoplasma pneumoniae pneumonia in mice’s lung tissue,reducing the apoptosis rate of lung tissue cells and expression of the TGF-β1 in lung tissue were part of its mechanism on Mycoplasma pneumoniae pneumonia.
