摘要
目的:基于升降浮沉药性理论,进行升浮药麻黄和沉降药葶苈子对风水水肿模型的作用比较,为中医“顺病位”用药原则提供数据支撑。方法:通过尾静脉注射阿霉素结合吹风刺激建立风水水肿模型。造模成功后,分别连续灌胃麻黄水煎液(4.67 mg/kg)和葶苈子水煎液(4.67 mg/kg)干预21 d。观察大鼠的基本状态,记录死亡率和脏器系数,并检测大鼠的中枢神经系统(自主活动次数、睡眠时间、旷场实验)、呼吸系统[诱咳实验、肺组织HE切片、BALF中的免疫球蛋白(IgE)、γ干扰素(IFN-γ)、白细胞介素(IL)-4水平]、泌尿系统[排尿量、足趾汗腺着色点数、肾组织病理、血清肌酐(CRE)、尿素氮(BUN)以及尿液中的尿蛋白(UP)和尿微量白蛋白(MAU)水平]、消化系统(小肠推进率、胃残留率、血浆胃动素(MTL)、生长抑素(SST)、胃泌素(GT)水平]以及循环系统(心功能、血清NO水平)的相关药理学指标以综合分析两者对风水水肿模型的影响。结果:两味药对呼吸和泌尿系统的作用显著,能不同程度改善肺部炎症及肾脏损伤,对循环系统无显著性影响;对于消化系统,麻黄表现出抑制作用,葶苈子表现出促进作用;此外,麻黄能够兴奋中枢(P<0.01),显著增加模型大鼠的足趾汗腺着色点数(P<0.01),降低血清中的BUN水平(P<0.05)。结论:升浮药麻黄治疗病位在表的风水水肿病症的综合效果显著优于沉降药葶苈子,符合“顺病位”用药原则,提示升降浮沉药性对特定方向或病势的病变起着显著或特殊的治疗作用。
Objective: Based on the theory of ascending and descending of traditional Chinese medicine, comparing the effects of the ascending medicine Ephedrae Herba and the descending medicine Descurainiae Semen Lepidii Semen on the wind edema model to provide data support for the principle of ‘following the location of the disease’ in traditional Chinese medicine. Methods: After the wind edema model was established by tail vein injection adriamycin combined with blowing stimulation, the Ephedrae Herba decoction(4.67 mg/kg) and the Descurainiae Semen Lepidii Semen decoction(4.67 mg/kg) were continuously administered for 21 days. Observe the basic state of rats, record mortality and organ coefficients, and detect the related pharmacological indicators of central nervous system(number of voluntary activities, sleep time, open field experiment),respiratory system(induced cough test, lung tissue pathology, the levels of IgE, IL-4 and IFN-γ in BALFL), urogenital system(urinary output, sweat output, renal tissue pathology, serum CER, BUN, and UP and MAU levels in urine), digestive system(small intestinal propelling rate, gastric residual rate, plasma MTL, GT, SST levels) and circulatory system(heart function, serum NO levels) to comprehensively analyze the effects of the two on the wind edema model. Results: The two herbs have significant effects on the respiratory and urinary systems, which can improve lung inflammation and kidney damage to varying degrees, without obvious effects on the circulatory system;For the digestive system, Ephedrae Herba has an inhibitory effect, and Descurainiae Semen Lepidii Semen has a promoting effect;In addition, Ephedrae Herba can excite nervous centrals(P<0.01), significantly increase the amount of perspiration in model rats(P<0.01), and reduce the level of BUN in the serum(P<0.05). Descurainiae Semen Lepidii Semen does not have these effects. Conclusion: The comprehensive therapeutic effect of the ‘ascending’ medicine Ephedrae Herba on wind edema model rats is better than that of the �
