LncRNA MEG3减轻缺血再灌注损伤所致肠黏膜屏障功能损害的机制

摘要 目的 探讨长链非编码RNA(Lnc RNA)MEG3减轻缺血再灌注肠黏膜屏障功能损害的机制,为肠缺血再灌注损伤的临床治疗提供潜在策略。方法 使用人结肠腺癌Caco-2细胞建立缺氧缺糖(OGD)模型。采用CKK-8法检测对照组、OGD处理组和OGD处理后合并Lnc RNA MEG3过表达或抑制表达后Caco-2细胞存活率,Annexin V-异硫氰酸荧光素(FITC)/碘化丙啶(PI)细胞凋亡检测试剂盒检测各组细胞凋亡,定量逆转录聚合酶链式反应检测各组细胞Lnc RNA MEG3表达的差异,采用跨皮细胞电阻(TEER)检测法测定肠黏膜TEER值,并分析肠屏障功能。结果 经OGD处理的各组Lnc RNA MGE3表达明显降低,细胞存活率降低且细胞凋亡增加,导入sh MEG3后进一步降低Caco-2细胞Lnc RNA MEG3表达及细胞存活率,并且加剧细胞凋亡,过表达Lnc RNA MEG3可增加OGD处理的各组Caco-2细胞存活率,减少其凋亡,并减轻其肠屏障功能损害。结论 Lnc RNA MEG3在体外实验中可减轻肠黏膜细胞缺血再灌注损伤,保护肠黏膜屏障功能。 Objective To explore the mechanism of the long non-coding RNA(Lnc RNA) MEG3 alleviating the functional damage of intestinal mucosal barrier after ischemia-reperfusion,and to provide potential strategies for the clinical treatment of intestinal ischemia-reperfusion injury.Methods The human colon adenocarcinoma Caco-2 cells were used to establish an oxygen and glucose deficiency(OGD) model.The CKK-8 assay was used to detect the survival rate of Caco-2 cells in the control group,OGD-treated group and after OGD treatment combined with Lnc RNA MEG3 overexpression or inhibition.The Annexin V-FITC/PI cell apoptosis detection kit was used to detect the apoptosis of different treatment groups.The quantitative reverse transcription polymerase chain reaction was used to detect the difference of Lnc RNA MEG3 expression in different treatment groups,and the transcutaneous cell resistance(TEER) was used to detect the intestinal mucosa TEER to analyze the intestinal barrier function.Results The results showed that the expression of Lnc RNA MEG3 decreased significantly,the cell survival rate decreased and the apoptosis increased in each group treated with OGD.The addition of sh MEG3 further reduced the expression of Lnc RNA MEG3 in Caco-2 cells,further reduced the cell survival rate and exacerbated the apoptosis.The addition of Lnc RNA MEG3 could increase the survival rate of Caco-2 cells,reduce their apoptosis and reduce the damage of intestinal barrier function in each group treated with OGD.Conclusion Lnc RNA MEG3 can reduce the ischemia-reperfusion injury of intestinal mucosal cells and protect the function of intestinal mucosal barrier in vitro.
