电针天枢、大肠俞对慢传输型便秘大鼠结肠组织ChAT、SCF和c-Kit表达的影响 被引量:6

摘要 目的观察电针“天枢”“大肠俞”对慢传输型便秘(STC)大鼠结肠组织乙酰胆碱转移酶(ChAT)、干细胞因子(SCF)和酪氨酸激酶受体c-Kit表达的影响,探讨电针治疗STC的作用机制。方法将40只SPF级SD大鼠随机分为空白组、盐水组、模型组和电针组,每组10只。模型组和电针组连续21 d灌胃复方地芬诺酯混悬液建立STC模型。造模成功后,电针组电针双侧“天枢”“大肠俞”,每日1次,连续14 d,其余各组固定相同时间,不干预。测定大鼠胃排空率和小肠推进率,免疫荧光检测大鼠结肠组织ChAT和c-Kit表达,Western blot检测结肠组织SCF、c-Kit蛋白表达,RT-PCR检测结肠组织SCF、c-Kit mRNA表达。结果与空白组比较,盐水组大鼠胃排空率、小肠推进率及结肠组织ChAT、SCF、c-Kit表达差异无统计学意义(P>0.05);与空白组比较,模型组大鼠胃排空率、小肠推进率显著降低(P<0.01),结肠组织ChAT和c-Kit表达显著降低,SCF、c-Kit蛋白及mRNA表达显著降低(P<0.01);与模型组比较,电针组大鼠胃排空率、小肠推进率显著升高(P<0.01),结肠组织ChAT、c-Kit表达显著升高,SCF、c-Kit蛋白及mRNA表达显著升高(P<0.01)。结论电针“天枢”“大肠俞”能改善STC大鼠胃肠道传输功能,其机制可能与促进ChAT表达/激活SCF/c-Kit通路有关。 Objective To observe the effects of electroacupuncture on“Tianshu”and“Dachangyu”on the expressions of ChAT,SCF,c-Kit in slow transit constipation(STC)rats;To explore the mechanism of electroacupuncture in the treatment of STC.Methods Totally 40 SD rats were randomly divided into blank group,saline group,model group and electroacupuncture group,with 10 rats in each group.STC rats model was established by intragastric administration of compound diphenoxylate suspension for 21 days.“Tianshu”and“Dachangyu”were selected as the acupoints in electroacupuncture group once a day for 14 consecutive days,the other groups were fixed for the same time without intervention.The gastric emptying rate and small intestinal propulsion rate were measured,immunofluorescence was used to detected the expressions of ChAT and c-Kit in colon tissue,Western blot was used to detected the expression of SCF and c-Kit protein expressions in colon tissue,RT-PCR was used to detected the expression of SCF and c-Kit mRNA in colon tissue.Results Compared with the blank group,there were no significant difference in gastric emptying rate,small intestinal propulsion rate,and the expression of ChAT,c-Kit,SCF of rats in the saline group(P>0.05).Compared with the blank group,the gastric emptying rate and small intestinal propulsion rate of rats in the model group significantly decreased(P<0.01),the expressions of ChAT and c-Kit in colon tissue significantly decreased,and the protein and mRNA expressions of SCF and c-Kit were significantly decreased(P<0.01).Compared with the model group,the gastric emptying rate and small intestinal propulsion rate of rats in the electroacupuncture group significantly increased(P<0.01),the expressions of ChAT and c-Kit in colon tissue significantly increased,and the protein and m RNA expressions of SCF and c-Kit significantly increased,with statistical significance(P<0.01).Conclusion Electroacupuncture on“Tianshu”and“Dachangyu”can improve the gastrointestinal transport function of STC rats,and its
