摘要
目的 探讨子宫内膜异位症患者经血和内膜间充质干细胞的表达和相关性。方法 选取2019年1—12月在舟山市妇女儿童医院就诊的子宫内膜异位症患者60例(异位组),同期健康体检结果正常育龄期妇女60例作为对照组。收集研究对象的月经血、子宫内膜组织(诊断性刮宫取得)、异位组患者异位内膜组织病灶子宫内膜组织(手术腹腔镜手术取得)。对标本体外细胞培养、分离及鉴定,以流式细胞仪检测间充质干细胞的表达。结果 子宫内膜间充质干细胞标志物CD29、CD44、CD90、CD105、CD146在对照组和异位组患者经血培养细胞中表达阳性率均超90.00%,差异无统计学意义(P>0.05),CD34和CD45表达阳性率低于10.00%,在异位组患者经血、在位内膜培养细胞中CD34和CD45表达阳性率高于对照组,差异有统计学意义(P<0.05),异位组患者经血、在位内膜培养细胞中CD34和CD45表达阳性率比较,差异无统计学意义(P>0.05)。结论 子宫内膜间充质细胞在经血、在位子宫内膜和异位子宫内膜间均有表达,但在子宫内膜异位症患者和正常人群中的表达有相似之处也有不同之处,子宫内膜异位症患者经血和在位内膜及异位内膜间充质干细胞表达表型并无明显不同。
Objective To discuss the expression and correlation of menstrual blood and endometrial mesenchymal stem cells in patients with endometriosis.Methods We collected 60 cases of patients with endometriosis(ectopic group) in Zhoushan Women’s and Children’s Hospital from January 2019 to December 2019,60 cases of women of childbearing age with normal physical examination results in the same period as the control group,to collect menstrual blood,endometrial tissue(diagnostic curettage collection),and lesion endometrial tissue of ectopic endometrium in the ectopic group(laparoscopic surgery collection),specimens of organizations were cultured in vitro,isolation and identification,the expression of mesenchymal stem cells was detected by flow cytometry.Results The positive rates of the markers CD29,CD44,CD90,CD105 and CD146 in the menstrual culture cells of the control group and the ectopic group were all over 90.00%,there was no statistically significant difference in the positive rate(P>0.05),the positive rates of CD34 and CD45 were lower than 10.00%,the positive rates of CD34 and CD45 expression were higher in the ectopic group than in the control group in menstrual blood and endometrial tissue cells,the difference was statistically significant(P0.05).Conclusion The endometrial mesenchymal stem cells were expression positive of menstrual blood and endometrial tissue cells,and lesion endometrial tissue cells of ectopic endometrium in patients with endometriosis,there are similarities and differences between endometriosis patients and the general population,the expression phenotypes of mesenchymal stem cells in different tissues were not significantly different in endometriosis patients.
