摘要
目的研究哮喘患儿T淋巴细胞中哺乳动物雷帕霉素靶蛋白(mTOR)信号通路活化及临床意义。方法选取我院收治的40例哮喘急性发作期患儿作为急性组,40例哮喘缓解期患儿作为缓解组,40例健康儿童作为健康组。比较三组的T淋巴细胞亚群、白细胞分化抗原4阳性(CD4^(+))T淋巴细胞亚群水平以及T淋巴细胞中p-Akt、p-mTOR、p-70S6K的表达水平。分别使用Spearman和Pearson检验分析mTOR信号通路活化与哮喘患儿病情和T淋巴亚群的相关性。结果急性组的白细胞分化抗原3阳性(CD3^(+))、白细胞分化抗原8阳性(CD8^(+))、辅助性T细胞1(Th1)、调节性T细胞(Treg)水平明显低于缓解组和健康组,CD4^(+)、辅助性T细胞2(Th2)、辅助性T细胞17(Th17)水平及p-Akt、p-mTOR、p-70S6K表达水平明显高于缓解组和健康组(P<0.05)。T淋巴细胞中p-Akt、p-mTOR、p-70S6K的表达与哮喘患儿病情、CD3^(+)、CD8^(+)、Th1、Treg均呈明显负相关,与CD4^(+)、Th2、Th17均呈明显正相关(P<0.05)。结论哮喘患儿T淋巴细胞中mTOR信号通路处于异常活化状态,并与哮喘患儿病情及T淋巴细胞亚群失衡明显相关。
Objective To study the activation of mammalian target of rapamycin(mTOR)signaling pathway in T lymphocytes of asthmatic children and its clinical significance Methods Forty children with acute attack of asthma admitted in our hospital were selected as acute group,40 children with asthma remission were selected as remission group,and 40 healthy children were selected as healthy group.The T lymphocyte subsets,cluster of differentiation 4 positive(CD4^(+))T lymphocyte subsets levels,and the expression levels of p-Akt,p-mTOR,p-70S6K in T lymphocytes were compared among the three groups.Spearman and Pearson tests were used to analyze the correlation between the activation of mTOR signaling pathway and the condition,T lymphocyte subsets in children with asthma.Results The levels of cluster of differentiation 3 positive(CD3^(+)),cluster of differentiation 8 positive(CD8^(+)),helper T cell 1(Th1),regulatory T cell(Treg)in the acute group were significantly lower than those in the remission group and the healthy group,while CD4^(+),helper T cell 2(Th2),helper T cell 17(Th17)levels,p-Akt,p-mTOR and p-70S6K expression levels were significantly higher than those in the remission group and the healthy group(P<0.05).The expression of p-Akt,p-mTOR and p-70S6K in T lymphocytes were significantly negatively correlated with the condition of children with asthma,CD3^(+),CD8^(+),Th1 and Treg,and significantly positively correlated with the CD4^(+),Th2 and Th17(P<0.05).Conclusion The mTOR signaling pathway in T lymphocytes of children with asthma is abnormally activated,and it is obviously related to the condition of children with asthma and the imbalance of T lymphocyte subsets.
