脑缺血再灌注损伤与线粒体凋亡的研究进展 被引量:8

摘要 脑梗死已成为严重的全球性疾病,常用治疗方法为疏通阻塞血管或恢复患病部位血液供应,但部分患者治疗后发生脑缺血再灌注损伤(CIRI),这导致脑梗死的治疗率下降,因此正确认识CIRI对于脑梗死的治疗至关重要。脑缺血再灌注后神经元的死亡与线粒体功能障碍所致的凋亡直接相关,ATP衰竭、活性氧大量堆积、氧自由基损伤、Ca^(2+)超载、线粒体通透性转换孔异常开放、线粒体膜电位异常、血脑屏障破坏、线粒体DNA受损、线粒体自噬等因素均可导致线粒体障碍,从而诱导线粒体凋亡通路。 Cerebral infarction has become a serious global disease.The common treatment is to dredge the blocked blood vessels or restore the blood supply of the diseased parts.However,some patients have cerebral ischemia-reperfusion injury(CIRI)after treatment,which leads to the decline of the treatment rate of cerebral infarction.Therefore,a correct understanding of CIRI is very important for the treatment of cerebral infarction.The death of neurons after cerebral ischemia-reperfusion is directly related to the apoptosis caused by mitochondrial dysfunction.ATP failure,large accumulation of reactive oxygen species,oxygen free radical damage,Ca^(2+) overload,abnormal opening of mitochondrial permeability transition pore,abnormal mitochondrial membrane potential,destruction of blood-brain barrier,damage of mitochondrial DNA,mitochondrial autophagy and other factors can all lead to mitochondrial dysfunction,thus inducing mitochondrial apoptosis pathway.
