摘要 通过对传统糖含量测定方法进行改进,将酶标仪检测技术引入本科生物化学实验教学中,用酶标仪代替紫外分光光度计对番茄汁中总糖含量进行测定,并对样品进行加标回收率实验研究。结果表明,酶标仪检测番茄汁中总糖含量为23.74 mg/mL,标准样标准偏差(SD)在0.001~0.014之间,相对偏差(CV)基本小于5%,样品SD为0.845,CV为1.8%,回收率在97%~110%之间,相对标准偏差(RSD)为4.52%,表明酶标仪测试数据具有良好的线性关系且精密度和准确性较好,达到良好的实验教学改革效果。 Microplate reader detection technology was introduced into the undergraduate biochemical experiment teaching process by improving the traditional sugar content determination method,the total sugar content in tomato juice was determined by microplate reader instead of ultraviolet spectrophotometer,and the recovery rate of samples was studied experimentally.The results show that the total sugar content in tomato juice is 23.74 mg/mL,the standard deviations of standard samples(SD)are 0.001-0.014,the relative deviation(CV)is less than 5%,with the SD of sample 0.845,CV is 1.8%,the recovery rates are 97%-110%,and the relative standard deviation(RSD)is 4.52%,which indicates that the test data using the microplate reader has a good linear relationship with good precision and accuracy,and achieves a good experimental teaching reform effect.
