维生素D_(3)对非酒精性脂肪性肝病大鼠血脂代谢的影响 被引量:1

Effects of vitamin D_(3)intervention on blood lipid metabolism in rats with nonal⁃coholic fatty liver disease

摘要 目的探讨维生素D_(3)干预对非酒精性脂肪肝大鼠体重、血脂、肝功的影响。方法高脂饮食喂养Wistar大鼠6 w建立非酒精性脂肪肝动物模型,随机分为对照组(CON组)、模型组(HFD组)、维生素D_(3)干预组(HFD+VD_(3)组)、溶媒对照组(HFD+Oil组)。维生素D_(3)干预7 w后采集血清及肝组织样本,测量体重、肝指数,采用全自动生化分析仪测定血清生化指标、肝功指标,HE染色观察肝组织病理学改变。结果相较于CON组,高脂饮食导致大鼠血清25(OH)D_(3)水平显著降低(P<0.05),肝指数增加(P<0.001),血清TG、TC、LDL-C水平均显著升高(P<0.05),HE染色显微镜下显示肝脏组织出现明显脂肪变性、气球样变、肝细胞小灶坏死伴炎细胞浸润。相比于HFD+Oil组,维生素D_(3)干预使得大鼠血清25(OH)D_(3)水平明显升高(P<0.05),肝指数显著降低(P<0.05);血清TG、TC、LDL-C、AST、ALT水平和肝组织TG、FFA水平均明显降低(P<0.001);肝脏组织脂肪病变程度有不同程度的降低。结论在剂量12.5μg/kg时,维生素D_(3)具有降低非酒精性脂肪肝大鼠的肝指数、纠正肝脏紊乱的脂代谢、修复肝细胞内皮损伤等积极作用。 Objective To investigate the effects of vitamin D_(3)intervention on body weight,blood lipid and liver function in rats with non-alcoholic fatty liver disease.Methods Wistar rats were fed with high-fat diet for 6 weeks to establish nonalcoholic fatty liver model,and then randomly divided into control group(CON group),model group(HFD group),vitamin D_(3)intervention group(HFD+VD_(3)group)and solvent control group(HFD+Oil group).After 7 weeks of intervention,serum and liver tissue samples were collected,body weight and liver index were measured,serum biochemical indexes and liver function indexes were determined by automatic bio⁃chemical analyzer,and liver histopathological changes were observed by HE staining.Results Compared with the CON group,the se⁃rum 25(OH)D_(3)level decreased significantly(P<0.05),liver index increased(P<0.001).Serum levels of TG,TC and LDL-C increased significantly(P<0.05).HE staining microscopy showed obvious steatosis,ballooning change,small focal necrosis of liver cells with in⁃flammatory cell infiltration.Compared with the HFD+Oil group,the serum 25(OH)D_(3)level increased significantly(P<0.05)and the liver index decreased significantly(P<0.05)after the intervention of vitamin D_(3).The levels of TG,TC,LDL-C,AST and ALT in serum and the levels of TG and FFA in liver tissue decreased significantly(P<0.001).The degree of adipose lesion in liver tissue decreased in different degrees.Conclusion Vitamin D_(3)at 12.5μg/kg had positive effects on reducing liver index,correcting liver metabolism dis⁃orders and repairing endothelial injuries of liver cells.
