双参芎连颗粒对动脉粥样硬化大鼠细胞凋亡的影响 被引量:3

摘要 目的观察双参芎连颗粒对动脉粥样硬化大鼠细胞凋亡的影响,探讨其改善动脉粥样硬化的作用机制。方法采用高脂饲料喂养及腹主动脉拉伤法造模,将大鼠分为空白组、模型组、丹蒌片组、舒降之组及双参芎连颗粒低、中、高剂量组,分别进行相应干预,连续8周。检测血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、氧化型低密度脂蛋白胆固醇(ox-LDL-C)、高敏C反应蛋白(hs-CRP)水平;HE染色观察腹主动脉组织病理变化;免疫组化染色腹主动脉组织检测Bcl-2、Bax蛋白表达。结果与空白组比较,模型组大鼠血清TC、LDL-C、ox-LDL-C、hs-CRP水平升高(P<0.01),腹主动脉组织可见较多炎性细胞浸润及大小不等的脂质空泡,斑块面积增加,腹主动脉组织Bcl-2、Bax蛋白表达显著升高(P<0.01),Bcl-2/Bax比值显著降低(P<0.01);与模型组比较,各给药组大鼠血清TC、LDL-C、ox-LDL-C、hs-CRP水平均不同程度降低,腹主动脉斑块内炎性细胞浸润减少,斑块面积缩小,腹主动脉组织Bcl-2蛋白表达显著升高(P<0.01),Bax蛋白表达显著降低,Bcl-2/Bax比值显著升高(P<0.01)。结论双参芎连颗粒能调节动脉粥样硬化大鼠脂质代谢,减轻炎症反应,抑制细胞凋亡,其作用机制与降低血清TC、LDL-C、ox-LDL-C、hs-CRP水平,上调Bcl-2蛋白表达,下调Bax蛋白表达有关。 Objective To observe the effects of Shuangshen Xionglian Granules on cell apoptosis in arteriosclerosis rats;To discuss its mechanism of improving arteriosclerosis.Methods The model was established by feeding high-fat diet and abdominal aorta strain.The rats were randomly divided into blank group,model group,Danlou Tablets group,simvastatin group,Shuangshen Xionglian Granules low-,medium-and high-dosage groups.Corresponding interventions were carried out for 8 consecutive weeks.The levels of total cholesterol(TC),triglyceride(TG),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),ox-LDL-C and hs-CRP were detected.The pathological changes in tissues of abdominal aorta were observed by HE staining,and immunohistochemical staining was used to detect the expressions of apoptosis-related proteins Bcl-2and Bax.Results Compared with the blank group,the levels of TC,LDL-C,ox-LDL-C and hs-CRP in serum of the rats in model group significantly increased(P<0.01),the abdominal aorta tissue showed more inflammatory cell infiltration and lipid vacuoles of different sizes,and the plaque area increased,the expressions of Bcl-2 and Bax protein in abdominal aorta tissue significantly increased(P<0.01),and the ratio of Bcl-2/Bax significantly decreased(P<0.01).Compared with the model group,the levels of TC,LDL-C,ox-LDL-C and hs-CRP in serum of the rats in each administration group decreased to varying degrees,inflammatory cell infiltration in abdominal aortic plaque was reduced,plaque area was reduced,the expression of Bcl-2 protein in abdominal aorta tissue significantly increased(P<0.01),the expression of Bax protein significantly decreased,and the ratio of Bcl-2/Bax significantly increased(P<0.01).Conclusion Shuangshen Xionglian Granules can regulate lipid metabolism in atherosclerotic rats,reduce inflammatory response,and inhibit apoptosis.The mechanism may be related to reducing serum TC,LDL-C,ox-LDL-C,hs-CRP levels,up-regulating Bcl-2 protein expression,down-regulating Bax protein expression
