摘要
目的 研究螺旋藻激酶对动脉粥样硬化大鼠血管的影响。方法 将SD大鼠随机分为6组:对照组、模型组、阳性药组和低、中、高剂量实验组。对照组大鼠给予常规饲料,其余组大鼠给予高脂饲料和维生素D3构建动脉粥样硬化大鼠模型,低、中、高剂量实验组分别灌胃给予80,160和320 U·kg^(-1)螺旋藻激酶溶液,阳性药组灌胃4 mg·kg^(-1)辛伐他汀混悬液,对照组和模型组灌胃给予等体积的0.9%NaCl溶液,每天给药1次,持续12周。以酶联免疫吸附法检测血清C反应蛋白(CRP)、基质金属蛋白酶9(MMP-9)的含量;以蛋白质印迹法检测腹主动脉的血管内皮生长因子(VEGF)蛋白的表达水平;以免疫组化法检测腹主动脉VEGF蛋白的表达水平;以苏木精-伊红染色法观察腹主动脉病理变化,并用Image Pro Plus软件测量其内膜-中膜厚度(IMT)。结果 对照组、模型组、阳性药组和低、中、高剂量实验组的CRP的含量分别(1 633.75±23.94),(2 177.50±145.06),(1 833.75±132.88),(1 971.50±37.67),(1 790.00±64.55),(1 608.75±89.85)ng·mL;MMP-9含量分别为(76.07±6.21),(109.70±17.91),(88.93±12.70),(92.86±14.97),(85.29±8.46),(81.43±6.78)ng·mL^(-1);IMT分别为(323.19±13.23),(492.79±6.41),(303.21±13.36),(348.99±5.55),(308.77±8.04),(288.00±6.02)μm。与模型组比较,对照组、阳性药组和中、高剂量实验组的CRP、MMP-9、IMT、VEGF指标,差异均有统计学意义(P<0.05或P<0.01)。结论 螺旋藻激酶对动脉粥样硬化大鼠血管有保护疗效,可能与降低炎症因子CRP、MMP-9和VEGF的表达水平有关。
Objective To explore effects of Spirulina kinase on vascular of atherosclerosis(AS) rats. Methods The SD rats were randomly divided into six groups: control group, model group, positive group, experimental-L,-M,-H groups. Control group rats were fed with normal diet, and the other groups were fed with high-fat diet and Vitamin D3 to establish atherosclerosis rat model. The experimental-L,-M,-H groups were given Spirulina kinase solution by gavage at the dose of 80, 160 and 320 U·kg^(-1)respectively;the positive group was given 4 mg·kg^(-1)simvastatin suspension by gavage, the control group and model group were given equal volume of 0.9% NaCl by gavage, and the rest were given relevant medicine by gavage, once a day,for consecutive 12 weeks. The serum levels of rat in C-reactive protein( CRP) and matrix metalloproteinase-9( MMP-9) were determined by enzyme-linked immunosorbent assay. Western blot was used to detect the expression level of vascular growth factor( VEGF) protein in rat abdominal aorta. The expression level of VEGF protein in rat abdominal aorta was detected by immunohistochemistry. The pathological changes of abdominal aorta in rats were observed by hematoxylin-eosin( HE) staining,and its intima-media thickness( IMT) was detected by Image Pro Plus software.
