摘要
目的探讨长链非编码RNA LINC00315在胃癌中的表达、临床意义及其对胃癌细胞增殖、凋亡的影响。方法选取2018年1月1日—2019年12月31日在宁波市医疗中心李惠利医院接受手术治疗并经术后病理诊断确诊的胃癌患者75例,收集每例患者的胃癌组织和对应的癌旁组织。胃癌细胞系(NCI-N87、SGC-7901、AGS、HCG27)和正常胃黏膜细胞(GES-1)均购自中国科学院上海细胞库。采用qRT-PCR法检测胃癌组织和各细胞系中LINC00315的表达水平,分析LINC00315表达水平与胃癌患者临床病理特征的关系。基于TCGA数据分析LINC00315的表达水平与胃癌患者预后的关系。采用MTT实验和EdU细胞增殖实验观察LINC00315对胃癌HCG27细胞增殖的影响;采用流式细胞术检测LINC00315对胃癌HCG27细胞凋亡的影响。结果LINC00315在胃癌组织中的表达水平较对应的癌旁组织升高(P<0.05),LINC00315在胃癌细胞系(NCI-N87、SGC-7901、AGS、HCG27)中的表达水平较正常胃黏膜细胞(GES-1)升高(P<0.05)。LINC00315表达水平与胃癌肿瘤大小及TNM分期有关(均P<0.05),高表达患者肿瘤较大、TNM分期较晚。TCGA数据分析结果显示:LINC00315高表达的胃癌患者总生存率、无病生存率均较低表达患者差(均P<0.05)。MTT实验和EdU细胞增殖实验表明:敲低LINC00315能够显著抑制胃癌HCG27细胞的增殖能力。流式细胞术分析结果显示:敲低LINC00315能够显著诱导胃癌HCG27细胞凋亡。结论LINC00315在胃癌中表达上调,可能是一个胃癌预后不良的潜在分子标志物,LINC00315能够促进胃癌细胞增殖,抑制胃癌细胞凋亡。
Objective To explore the expression and clinical significance of LINC00315 in gastric cancer(GC)and its effect on the proliferation and apoptosis of GC cells.Methods Seventy-five patients with gastric cancer who underwent surgical treatment and were diagnosed by postoperative pathology at Ningbo Medical Center Lihuili Hospital from January 2018 to December 2019 were enrolled,and gastric cancer tissues and corresponding were collected from each patient.LINC00315 expression was detected by qRT-PCR in cancer tissues and paricancerous tissues of GC patients,and also in GC cell lines NCI-N87,SGC-7901,AGS,HCG27 and normal gastric mucosal cells GES-1.The correlation between LINC00315 expression and clinicopathological features of GC was analyzed.The association between LINC00315 expression and the prognosis of GC patients was analyzed using TCGA data.MTT assay and EdU assay were employed to explore the effect of LINC00315on GC cell proliferation.Flow cytometry analysis was applied to detect the effect of LINC00315 on the apoptosis of GC cells.Results LINC00315expression was significantly increased in GC tissues compared to adjacent gastric mucosal tissues(P<0.05).LINC00315level was significantly increased in GC cell lines(NCI-N87,SGC-7901,AGS,HCG27)compared to normal gastric mucosal cells(GSE-1)(P<0.05).LINC00315 expression was significantly associated with tumor size and TNM stage(P<0.05),and patients with high expression showed larger tumors and advanced TNM stage.TCGA data analysis revealed that GC patients with higher LINC00315 expression had worse overall survival and disease free survival rates(P<0.05).MTT assay and EdU assay revealed that LINC00315 knockdown significantly inhibited the proliferation of GC cells.Meanwhile,flow cytometry analysis showed that GC cell apoptosis was induced by LINC00315 knockdown.Conclusion LINC00315 expression is upregulated in GC,indicating that it may be a potential molecular marker for predicting the poor prognosis of GC.LINC00315 promotes the cell proliferation and suppresses apo
