雪白睡莲花总黄酮提取工艺优化及其对金黄色葡萄球菌性小鼠乳腺炎模型的治疗效果 被引量:5

Optimal extraction process and therapeutic effect studies of total flavonoids from Nymphaea candida on mice model of Staphylococcus aureus mastitis

摘要 为探索响应面法结合超声辅助提取雪白睡莲花总黄酮的最优工艺条件。本试验以雪白睡莲花为材料,在单因素试验的基础上,采用响应面法考察乙醇浓度、超声功率、提取时间对雪白睡莲花总黄酮得率的影响;采用牛津杯法结合倍比稀释法测定抑菌圈、最低抑菌浓度(MIC)及最低杀菌浓度(MBC),研究雪白睡莲花总黄酮对金黄色葡萄球菌在体外的抑制效果;同时,构建金黄色葡萄球菌性小鼠乳腺炎模型,定期检测各试验组小鼠乳腺组织中金黄色葡萄球菌的数量、TNF-α及IL-6的含量,观察小鼠乳腺组织病理学变化,研究雪白睡莲花总黄酮对金黄色葡萄球菌性小鼠乳腺炎模型的体内治疗效果。结果显示,雪白睡莲花总黄酮最佳提取工艺条件为:乙醇浓度81%,超声功率60%,提取时间20 min,料液比1∶30 kg/L,提取温度60℃。在此条件下,雪白睡莲花总黄酮得率达到12.00%,与预测值11.99%相近。与小鼠乳腺炎模型组对比,雪白睡莲花总黄酮组小鼠乳腺组织中金黄色葡萄球菌的数量、TNF-α及IL-6的含量均显著降低(P<0.01),乳腺组织中的炎症细胞和红细胞明显减少。结果表明,采用响应面法对雪白睡莲花总黄酮提取条件进行优化合理可行,雪白睡莲花总黄酮具有较好的体内抑菌抗炎功效。 The purpose of this experiment was to study the optimal extraction conditions for the preparation of total flavonoids from Nymphaea candida by response surface methodology combined with ultrasonic assisted extraction.Nymphaea candida was used as the material, by single factor experiment, the effect of ethanol concentration, ultrasonic power, and extraction time on the extraction rate of total flavonoids from Nymphaea candida was studied with response surface methodology.The inhibition zone, minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) of total flavonoids from Saussurea involucrata on Staphylococcus aureus in vitro were determined by Oxford cup method and multiple dilution method.In addition, the antibacterial and anti-inflammatory effects of total flavonoids from Nymphaea candida in vivo were explored through the establishment of a mice model of Staphylococcus aureus mastitis.Colony-forming unit(CFU) of Staphylococcus aureus,the content of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) in breast tissue were detected, and the histopathological section of breast was made.The results showed that the optimal extraction process conditions for total flavonoids from Nymphaea candida were as follows:best concentration of ethanol was 81%,ultrasound power was 60%,ultrasound treatment time was 20 min,a ratio of ethanol to liquid was 1∶30 and temperature of extraction was 60℃.Under these conditions,the measured value of total flavonoids from Nymphaea candida was 12.00%,which was close to the predicted value of 11.99%.Compare with the mice model group of Staphylococcus aureus mastitis,the CFU of Staphylococcus aureus,the content of TNF-αand IL-6 in breast tissue of total flavonoids group from Nymphaea candida was significantly decreased(P<0.01),inflammatory cells and erythrocytes were significantly reduced in the breast tissue.In conclusion,it was feasible to adopt the response surface methodology to optimize the extraction conditions of flavonoids,and the total flavonoids f
