金银花、连翘对金黄色葡萄球菌诱发小鼠乳腺炎的防治作用研究 被引量:8

摘要 目的:研究中草药方剂预防和治疗小鼠乳腺炎炎症模型的效果。方法:用金黄色葡萄球菌感染小鼠建立乳腺炎炎症模型,后灌服金银花、连翘及其复方水煎液,观察其治疗乳房炎的效果;正常小鼠给药后,用金黄色葡萄球菌感染,观察其预防乳腺炎的效果。用平板计数法计数各组乳腺组织的细菌感染数量;制作乳腺组织的石蜡切片,观察H.E染色和甲苯胺染色的乳腺组织病理变化、肥大细胞的数量和分布部位;尾静脉采集各组小鼠的血液,用ELISA法检测血清中TNF-α的含量。结果:预防组、治疗组与模型组比较,小鼠乳腺中细菌感染的数量均减少,且金银花的作用较连翘和混合药液显著;观察乳腺组织及细胞发现,预防组和治疗组对小鼠乳腺组织的病理变化均有一定的改善作用;预防组和治疗组小鼠血清中TNF-α的含量较模型组均降低。结论:金银花、连翘及其复方药液对小鼠乳腺炎均有很好的治疗效果,对该病原菌引起的乳房炎有较好的预防效果,其中金银花的作用效果最显著。 Objective: To study the preventive and therapeutic effects of Flos Lonicerae and Fructus Forsythiae on mice mastitis induced by Staphylococcus aureus. Methods: The mastitis model was established with mice infected with Staphylococcus aureus. Then the mice were treated by the decoction of Flos Lonicerae, Fructus Forsythiae or their combined preparation, respectively. Healthy mice were challenged after given the medicine to study the preventive effect. Plate count method was used to count the number of Staphylococcus aureus in each group of breast tissue. Breast tissue was sliced into paraffin sections, stained with H.E and toluidine blue, and observed the pathological changes and the number of mast cell as well as the position they distributed in breast tissue by light microscope. The serum was separated from the blood in tail vein to detect the content of TNF-α. Results: The infected bacteria number in Chinese medicine preventive group and the treatment group were all reduced, and Flos Lonicerae treated group present the most significant effect in the three groups. Microscopic observation showed that there were some improvement in the pathological changes of preventive group and treatment group,meanwhile, the TNF-α content of them were decreased compared with the model group. Conclusion: Flos Lonicerae, Fructus Forsythiae and their combined preparation has curative and preventive effect on mice mastitis induced by Staphylococcus aureus, and Fructus Forsythiae present the most significant activity.
