基于多个miRNAs调节NF-κB信号通路探讨艾灸干预腹泻型肠易激综合征模型大鼠的抗炎机制研究 被引量:11

Anti-inflammation effect of moxibustion for rats with diarrhea-predominant irritable bowel syndrome based on multiple miRNAs regulating NF-κB signal pathway

摘要 目的:观察艾灸对腹泻型肠易激综合征(IBS-D)模型大鼠多个微小RNAs(miRNAs)调节核因子-κB(NF-κB)、炎性因子的影响,探讨艾灸干预IBS-D的抗炎机制。方法:从52只初生幼鼠中随机抽取12只作为正常组,剩余大鼠制备IBS-D模型,将36只造模成功的大鼠随机分为模型组、药物组和艾灸组,每组12只。药物组予腹腔注射吡咯烷二硫代甲酸铵盐(PDTC);艾灸组予“天枢”“上巨虚”艾灸治疗,每次20 min,均每天1次,共干预7 d。于造模前后及干预后测量各组大鼠体质量、稀便率和腹部回缩反射(AWR)最小容量阈值;干预后采用ELISA法检测各组大鼠血清肿瘤坏死因子α(TNF-α)和白细胞介素(IL)-1β、IL-8含量,HE染色观察各组大鼠结肠组织形态,实时荧光定量PCR法检测大鼠结肠组织miR-155、miR-125b、miR-29b、miR-31、miR-18a及NF-κB p65 mRNA的表达量,免疫荧光法检测大鼠结肠组织NF-κB p65、TNF-α、IL-1β、IL-8的蛋白表达。结果:造模后模型组大鼠体质量、AWR最小容量阈值均低于正常组(P<0.01),稀便率均高于正常组(P<0.01);干预后,模型组大鼠结肠组织炎性细胞浸润明显,血清TNF-α、IL-1β、IL-8含量均高于正常组(P<0.05),结肠组织miR-155、miR-125b、miR-29b、miR-31、miR-18a及NF-κB p65mRNA表达量均高于正常组(P<0.05),结肠组织NF-κB p65、TNF-α、IL-1β、IL-8蛋白表达亦高于正常组(P<0.01)。干预后,药物组和艾灸组大鼠体质量、AWR最小容量阈值均高于模型组(P<0.05),稀便率均低于模型组(P<0.05),结肠组织炎性细胞浸润较模型组减少,血清TNF-α、IL-1β、IL-8含量及结肠组织NF-κB p65、TNF-α、IL-1β、IL-8蛋白表达均低于模型组(P<0.05,P<0.01);药物组大鼠结肠组织miR-125b、miR-31、miR-18a及NF-κB p65 mRNA表达量均低于模型组(P<0.05),艾灸组大鼠结肠组织miR-155、miR-125b、miR-29b、miR-31、miR-18a及NF-κB p65 mRNA表达量均低于模型组(P<0.05)。大鼠结肠组织miR-155、miR-125b� Objective To observe the effect of moxibustion on the regulation of nuclear factor-kappa B(NF-κB)and inflammatory factors by multiple microRNAs(miRNAs)in rats with diarrhea-predominant irritable bowel syndrome(IBS-D),and to explore the anti-inflammatory mechanism of moxibustion on IBS-D.Methods Twelve of 52 newborn rats were randomly selected into a normal group.The remaining rats were made into IBS-D model.A total of 36 rats with successful model were randomly divided into a model group,a medication group and a moxibustion group,12 rats in each group.The rats in the medication group were intraperitoneally injected with pyrrolidine dithiocarbamate(PDTC).The rats in the moxibustion group were treated with moxibustion at"Tianshu"(ST 25)and"Shangjuxu"(ST 37)for 20 min each time.All the intervention was given once a day for 7 days.Before and after modeling as well as after intervention,the body mass,loose stool rate and the minimum volume threshold of abdominal withdrawal reflex(AWR)were measured.After intervention,the contents of serum tumor necrosis factorα(TNF-α),interleukin(IL)-1βand IL-8 were detected by ELISA method;the morphology of colon tissues was observed by HE staining,and the expressions of miR-155,miR-125 b,miR-29 b,miR-31,miR-18 a and NF-κB p65 mRNA in colon tissues were detected by real-time PCR.The expressions of NF-κB p65,TNF-α,IL-1βand IL-8 protein in colon tissues were detected by immunofluorescence.Results After modeling,the body mass and the minimum volume threshold of AWR in the model group were lower than those in the normal group(P<0.01);the rates of loose stool in the model group were higher than those in the normal group(P<0.01);after intervention,in the model group,the inflammatory infiltration of colon tissues was obvious,and the serum levels of TNF-α,IL-1β,IL-8 were higher than those in the normal group(P<0.05);the expression of miR-155,miR-125 b,miR-29 b,miR-31,miR-18 a and NF-κB p65 mRNA in colon tissues was higher than that in the normal group(P<0.05);the protein expression
