摘要
目的:探讨亚甲基四氢叶酸还原酶(MTHFR)基因C677T位点多态性与精神分裂症伴抑郁的关系。方法:采用卡尔加里精神分裂症抑郁量表(CDSS)对413例稳定期精神分裂症患者(病例组)进行抑郁症状评估,并分为伴抑郁亚组(n=198)与不伴抑郁亚组(n=215)。采用阳性与阴性症状量表(PANSS)评估患者的精神症状,使用聚合酶链反应-限制性片段长度多态性方法(PCR-RFLP)进行C677T位点基因多态性分析;结果与452名健康对照者(对照组)比较。比较两亚组间MTHFR基因C677T位点基因型及等位基因频率。结果:伴抑郁亚组T等位基因频率明显高于不伴抑郁亚组(P<0.05)。PANSS评分在病例组不同基因型患者间差异无统计学意义。CDSS评分在不同基因型患者间差异具有统计学意义,TT基因型的患者CDSS评分显著高于CT或CC基因型患者(P<0.05)。结论:MTHFR基因与精神分裂症患者伴发抑郁之间存在相关性,C677T位点TT基因型携带者可能与抑郁症状的发生有关。
Objective:To explore the relationship between methylenetetrahydrofolate reductase(MTHFR)gene C677T polymorphism and schizophrenia with depression.Method:Depressive symptoms of 413 stable schizophrenia patients(case group)were assessed with Calgary Depression Scale for Schizophrenia(CDSS).Patients were diveded into depression subgroup(n=198)and non-depression subgroup(n=215).The psychiatric symptoms were assessed by Positive and Negative Symptom Scale(PANSS),and the C677T polymorphism was genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),and the results were compared with 452 healthy controls(control group).MTHFR gene C677T genotype and allele frequency were compared between the two subgroups.Results:The frequency of T allele in the depression subgroup was significantly higher than that in the non-depression subgroup(P<0.05).There was no significant difference in PANSS score among patients with different genotypes.There was significant difference in CDSS score among patients with different genotypes and the CDSS score of patients with TT genotype was significantly higher than those with CT or CC genotype(P<0.05).Conclusion:MTHFR gene is associated with schizophrenia with depression,and C677T polymorphism TT genotype may be associated with the development of depressive symptoms.
