急性痛风性关节炎小鼠模型制备方法的改良 被引量:7

摘要 目的:建立一种改良的急性痛风性关节炎(acute gouty arthritis,AGA)小鼠模型并与传统制备方法比较。方法:制备单钠尿酸盐(monosodium urate,MSU)晶体干燥保存,30只8周龄雄性C57BL/6小鼠法随机分为传统方法造模组、改良方法造模组和对照组每组各10只,使用0.3 mL注射器向小鼠右踝关节注射0.9%氯化钠溶液设为对照组。传统方法制备AGA模型,研磨MSU晶体用0.9%氯化钠溶液配成MSU晶体混悬液(50 mg•mL^(-1)),采用27号针头的1 mL注射器沿小鼠右踝关节外侧后方垂直穿刺进入踝关节腔注入30μL MSU混悬液。改良方法制备AGA模型:MSU晶体研磨后,再用40μm细胞过滤器过筛,配成相同浓度MSU晶体混悬液,采用30号针头的0.3 mL注射器同法制备模型,于造模前和造模后不同时间点测量各组小鼠右侧踝关节直径,造模7 d后取材、HE染色观察各组小鼠踝关节组织病理变化。结果:MSU晶体镜下呈针尖状聚集成核,研磨的MSU晶体颗粒变小,但大小不均匀;经过细胞过滤器筛过的MSU晶体颗粒较为均匀、直径小于40μm。与对照组比较,2种造模方法制备的模型小鼠在造模6、12、18和24 h右踝关节肿胀程度均显著升高(P<0.01),其中传统方法造模小鼠在6、12、18和24 h右踝关节肿胀程度较改良方法造模小鼠更明显(P<0.05),但组内肿胀程度差异很大,改良方法造模小鼠组内肿胀程度差异较小。HE染色观察到,传统方法造模小鼠右踝关节滑膜组织显著增生并侵犯骨质,造成骨破坏,改良方法造模小鼠关节滑膜组织明显增生但软骨平滑完整无损伤。结论:改良的小鼠AGA模型造模方法稳定性优于传统造模方法,可用于单一痛风疾病的机制研究、抗痛风药疗效的筛选及其作用机制的研究,是一种可靠的动物模型。 Objective:To establish an improved acute gouty arthritis(AGA)mouse model and compare the improved method with traditional method.Methods Preparing monosodium urate(MSU)crystals and dried for storage.In the present study,308-week-old male C57BL/6 mice were randomly divided into traditional preparation group,improved preparation group,and control group,and each group contained 10 mice.Using a 0.3 mL syringe to inject 0.9%sodium chloride solution into the right ankle joint of the mouse as the control group.Preparation of mouse AGA model with traditional method:Grind MSU crystals,prepare MSU crystal suspension(50 mg•mL^(-1))with 0.9%sodium chloride solution,a 1mL syringe with 27-gauge needle was used to puncture vertically into the ankle cavity along the lateral rear of the mouse's right ankle joint,and inject 30μL MSU suspension.Preparation of mouse AGA model with improved method:After grinding the MSU crystals,sieving them with a 40μm cell strainer to prepare a suspension of MSU crystals of the same concentration,and prepare the model with a 0.3 mL syringe with 30-gauge needle.The diameter of the right ankle joint was measured at different time points before and after the modeling.The samples were taken 7 days after the modeling,and HE staining was used to observe the histopathological changes of ankle joint of mice in each group.Results:After preparation,the MSU lens showed a pinpoint shape,gathered into a nucleus.The ground MSU crystal particles became smaller,but the sizes were not uniform.After sieved through the cell strainer,MSU crystal particles became relatively uniform,the average diameter was less than 40μm.Compared with the control group,the swelling degree of the right ankle joint of the model mice prepared by the two modeling methods increased significantly at 6,12,18,and 24 hours after modeling(P<0.01).The swelling degree of the right ankle joint of the mice modeled by the traditional method was more obvious at 6,12,18 and 24 h than the mice modeled by the modified preparation method(P<0.05).Howe
